Periodontitis is a chronic inflammatory disease induced by bacteria. Exposure of the host to periodontal pathogens and their virulence factors induces a state of hyporesponsiveness to subsequent stimulations, which is termed endotoxin tolerance. The role and mechanism of lipopolysaccharide (LPS)-tolerized monocytes in inflammatory responses in neutrophils are currently unclear. Here, conditione...

The aim of this work was to analyze the effect of Interferon(IFN) and tumor necrosis factor(TNF) on NADPH oxidase activity and gp91-phox gene expression in human colostrum macrophages (CM), peripheral blood monocytes (PBM), and myelomonocytic THP-1 cells. We also investigated the effect of IFNon the release of TNFby these cells. Our results show that under basal culture conditions, CM release m...

Fifteen monoclonal antibodies have been produced to human Tamm-Horsfall protein (THP), identifying at least seven distinct epitopes. The antibodies have been used to isolate from serum an immunoreactive protein which comigrates with urinary THP. In addition, the antibodies may prove useful to set up an immunoenzymoassay for urinary THP as well as for immunoaffinity purification.

Macrophage colony-stimulating factor (M-CSF) is required for the proliferation, differentiation, and activation of monocytes. High-affinity receptors for M-CSF are encoded by the c-fms proto-oncogene. In the present study, we show that c-fms transcripts are detectable in human THP-1 myeloid leukemia cells. Furthermore, radiolabeled 125I-M-CSF is rapidly internalized into THP-1 cells and then de...

Tamm-Horsfall protein (THP) is a glycoprotein uniquely expressed in the kidney. We recently showed an important role for THP in mediating tubular cross-talk in the outer medulla and in suppressing neutrophil infiltration after kidney injury. However, it remains unclear whether THP has a broader role in neutrophil homeostasis. In this study, we show that THP deficiency in mice increases the numb...

A null cell leukemia cell line, THP-5, was established from the peripheral blood of a patient with acute null cell leukemia. THP-5 cells were characterized by the presence of Ia-like antigen and the absence of other cell surface markers examined. Despite the presence of Ia like antigen, THP-5 cells had no stimulating capacity in autologous or allogeneic mixed lymphocyte reaction.

Suspension cultures of a human monocytic leukemia cell line, THP-1, were treated with 0.16 to 160 nw 12-O-tetradecanoylphorbol-13-acetate (TPA). In an original cell line, THP1-O, cultured again from —80°cryopreservation, more than 80% of the cells adhered to the glass substrate with marked morphological change within 3 hr of TPA treatment. Adherent cells became flat and amoeboid in shape, an...

Neurosteroids are a class of steroids synthesized de novo in the brain, several of which are potent modulators of GABA(A) receptor function. In developing brain GABA(A) receptor, stimulation plays a trophic role. Cortical levels of the GABAergic neurosteroid 3alpha-hydroxy-5alpha-pregnan-20-one (3alpha,5alpha-THP) vary dramatically across development; during the second week of life, elevated le...

In the present study we investigated whether endothelial microparticles (EMPs) can bind to monocytic THP-1 cells and modulate their procoagulant properties. Using flow cytometry, we demonstrated that EMPs express adhesive receptors similar to those expressed by activated endothelial cells. Expression of endothelial antigens by THP-1 cells incubated with EMP was shown by immunoperoxidase stainin...
