In this paper we introduce a new variant of splicing systems, called weighted splicing systems, and establish some basic properties of language families generated by this type of splicing systems. We show that a simple extension of splicing systems with weights can increase the computational power of splicing systems with finite components.

The four Hu [embryonic lethal abnormal vision-like (ELAVL)] protein family members regulate alternative splicing by binding to U-rich sequences surrounding target exons and affecting the interaction of the splicing machinery and/or local chromatin modifications. Each of the Hu proteins contains a divergent N-terminus, three highly conserved RNA recognition motifs (RRM1, RRM2 and RRM3) and a hin...

Objective(s): The use of antisense oligonucleotides (AOs) to restore normal splicing by blocking the recognition of aberrant splice sites by the spliceosome represents an innovative means of potentially controlling certain inherited disorders affected by aberrant splicing. Selection of the appropriate target site is essential in the success of an AO therapy. In this study, in search for a splic...

Figure S1. Additional validations of microarray predicted ESRP regulated splicing events. Semiquantitative RT-PCR was used to assay a set of more complex candidate alternative splicing events. Identities of PCR products are labeled to the right of the gels. The percent inclusion of the relevant exon(s), shaded in red, is given below each lane. For the mutually exclusive splicing events in the F...

The polymerase chain reaction is fundamental to molecular biology and is the most important practical molecular technique for the research laboratory. We have developed and tested efficient tools for PCR primer and probe design, which also predict oligonucleotide properties based on experimental studies of PCR efficiency. The tools provide comprehensive facilities for designing primers for most...

Alternative splicing enables a single gene to produce multiple mRNA isoforms by varying splice site selection. In animals, alternative splicing of mRNA isoforms between cell types is widespread and supports cellular differentiation. In plants, at least 20% of multi-exon genes are alternatively spliced, but the extent and significance of tissue-specific splicing is less well understood, partly b...

Molecular cloning methods based on primer and overlap-extension PCR are widely used due to their simplicity, reliability, low cost and high efficiency. In this article, an efficient mega primer-mediated (MP) cloning strategy for chimaeragenesis and long DNA fragment insertion is presented. MP cloning is a seamless, restriction/ligation-independent method that requires only three steps: (i) the ...

background human telomerase reverse transcriptase (htert) has become an ideal target for development of anticancer therapy. small interfering rnas (sirnas) are very powerful reagents for gene silencing and show promise for cancer gene therapy. however, only a small number of sirnas have been demonstrated to be effective. for gene therapy targeting htert, it is essential to develop a robust syst...

BACKGROUND Accurate evaluation of unclassified sequence variants in cancer predisposition genes is essential for clinical management and depends on a multifactorial analysis of clinical, genetic, pathologic, and bioinformatic variables and assays of transcript length and abundance. The integrity of assay data in turn relies on appropriate assay design, interpretation, and reporting. METHODS W...
