Studies investigating the feasibility of new, or improved, biotechnologies, such as wastewater treatment digesters, inevitably start with laboratory-scale trials. However, it is rarely determined whether laboratory-scale results reflect full-scale performance or microbial ecology. The Expanded Granular Sludge Bed (EGSB) bioreactor, which is a high-rate anaerobic digester configuration, was used...

Obtaining a reliable estimation of the methane potential of organic waste streams in anaerobic digestion, for which a biochemical methane potential (BMP) test is often used, is of high importance. Standardization of this BMP test is required to ensure inter-laboratory repeatability and accuracy of the BMP results. Therefore, guidelines were set out; yet, these do not provide sufficient informat...

An up-flow anaerobic sludge blanket (UASB) reactor targeting sulfate reduction was operated under a constant TOC/S-SO42− ratio of 1.5 ± 0.3 g C/g S for 639 days using crude glycerol as carbon source. A filamentous and fluffy flocculant material, namely slime-like substances (SLS), gradually accumulated in the bioreactor after cease methanogenic activity. The accumulation SLS followed by decreas...

Atrazine degradation by anaerobic mixed culture microorganism in co-metabolic process and in absence of external carbon and nitrogen source was studied at influent atrazine concentration range of 0.5-15 mg/l. Wastewater of desired characteristic was prepared by the addition of various constituents in distilled water spiked with atrazine. In co-metabolic process, dextrose of various concentratio...

objective(s) some properties of neuraminidase produced by pseudomonas aeruginosa pao1 growth in a defined medium (bhi) were examined and evaluated for its features. materials and methods the obtained supernatant enzyme of p. aeruginosa pao1 cultures was used in a sensitive fluorometric assay by using 2'-(4-methylumbelliferyl) a-d-n acetylneuraminic acid as substrate. as hydrolyzing mun wit...

This study aims to investigate thermophilic methanogens in turf used as an inoculum. Results showed that Methanoculleus sp. regarded as hydrogenotrophic and Methanosarcina sp. regarded as acetoclastic methanogens were present in turf tested. However, active acetoclastic methanogens were present in turf soil only. The current study showed that thermophilic methanogens were present in various tur...

Marine sponges are natural sources of brominated organic compounds, including bromoindoles, bromophenols, and bromopyrroles, that may comprise up to 12% of the sponge dry weight. Aplysina aerophoba sponges harbor large numbers of bacteria that can amount to 40% of the biomass of the animal. We postulated that there might be mechanisms for microbially mediated degradation of these halogenated ch...

Despite its global abundance, Bacillus pumilus is poorly studied. The Fairview strain was obtained from a methanogenic anaerobic coal digester. The draft genome sequence was 3.8 Mbp long and contained 3,890 protein-coding genes. Like the SAFR-032 strain, it includes B. pumilus-specific proteins that likely confer enhanced resistance to environmental stresses.

We examined the survival potential of methanogenic archaea exposed to different environmental stress conditions such as low temperature (down to -78.5 degrees C), high salinity (up to 6 M NaCl), starvation (up to 3 months), long-term freezing (up to 2 years), desiccation (up to 25 days) and oxygen exposure (up to 72 h). The experiments were conducted with methanogenic archaea from Siberian perm...

Methanogenic archaea produce methane as a metabolic product under anoxic conditions and they play a crucial role in the global methane cycle. In this study molecular diversity of methanogenic archaea in the hyporheic sediment of the lowland stream Sitka (Olomouc, Czech Republic) was analyzed by PCR amplification, cloning and sequencing analysis of the methyl coenzyme M reductase alpha subunit (...
