Freshly-harvested seeds of Avena sativa L. do not germinate when imbibed at temperatures higher than 25 degrees C. This high temperature dormancy is due to the seed coats, and to the low activities of glycolysis and the oxidative pentose phosphate pathway (OPP) in the embryo. The analysis by exclusion chromatography of soluble NADP(+) phosphatase activities of embryos revealed two isoforms: a 3...

Abstract The gases used: ethylene (C 2 H 4 ), hydrogen cyanide (HCN) and nitric oxide (NO) showed a high activity as inductors of germination in primary dormant or non-dormant seeds exposed to stress suboptimal temperatures. So far, research on the role ethylene, has involved these during seed germination. This work describes gas-priming novel method for treating air dry genus Amaranthus servin...

Seed coats of six pea genotypes contrasting in dormancy were studied by laser desorption/ionization mass spectrometry (LDI-MS). Multivariate statistical analysis discriminated dormant and non-dormant seeds in mature dry state. Separation between dormant and non-dormant types was observed despite important markers of particular dormant genotypes differ from each other. Normalized signals of long...

We investigated the sensitivity of dormant Polish-provenance Fraxinus excelsior L. seeds to extreme desiccation and liquid nitrogen (LN, -196 degrees C). Germination and seedling emergence tests revealed that the critical water content (WC) of the desiccated seeds was 0.04-0.06 g H(2)O g(-1) dry mass, g g(-1) (0.04 g g(-1) in the germination test and 0.06 g g(-1) in the seedling emergence test)...

Setaria viridis has recently emerged as a promising genetic model system to study diverse aspects of monocot biology. While the post-germination life cycle of S. viridis is approximately 8 weeks long, the prolonged dormancy of freshly harvested seeds can more than double the total time required between successive generations. Here we describe methods that promote seed germination in S. viridis....

Plant seeds will not germinate when in a dormant state. What then are the factors controlling dormancy? A recent study has uncovered two basic-helix-loop-helix proteins which repress germination and are critical for maintaining dormancy. The combined action of light and cold synergistically alleviates activity of these repressors thus stimulating germination.

An enzymatically active recombinant protein kinase, previously isolated and characterized in Fagus sylvatica L. dormant seeds (FsPK1), was used to obtain a specific polyclonal antibody against this protein. Immunoblotting and immunohistochemical analysis of FsPK1 protein in beech seeds showed a strong immunostaining in the nucleus of the cells located in the vascular tissue of the embryonic axi...

Radicles (1 mm long) of pea embryo axes, excised from dormant seeds and from seeds soaked for 80 min, 8, 12, and 48 hr, were fixed in glutaraldehyde and postfixed in OsO(4). The nucleus was only slightly changed by 48 hr soaking, but cytoplasmic organelles underwent developmental changes. There was proliferation of endoplasmic reticulum, the appearance of dictyosomes, and an inward migration of...
