We demonstrate a simple and cost-efficient scanning confocal microscope setup for use in advanced instructional physics laboratories. The is constructed from readily available commercial products, the implementation of 3D-printed flexure stage allows further cost reduction pedagogical opportunity. Experiments exploring thickness slide surface solid objects with height variation are presented as...

We propose a new type of confocal microscope using Fresnel incoherent correlation holography (FINCH). Presented here is a confocal configuration of FINCH using a phase pinhole and point illumination that is able to suppress out-of-focus information from the recorded hologram and hence combine the super-resolution capabilities of FINCH with the sectioning capabilities of confocal microscopy.

Wide-field fluorescence microscopy is a commonly used imaging technique by researchers and clinicians. A standard wide-field microscope has no optical sectioning capabilities and this limits its use in imaging thick biological samples. Although standard wide-field fluorescence microscopy with deconvolution techniques can improve image quality [1], it does not provide true optical sectioning, du...

The Confocal Laser Scanning Microscope (CLSM) is an optical microscope that includes a laser light as a light source and an electronic system which helps on image processing. It obtains high-resolution and extremely thin optical image sections, removing the interference caused by the light arriving from the different optical fields across the thickness of the sample, and focusing on a single pl...

Zebrafish have long been utilized to study the cellular and molecular mechanisms of development by time-lapse imaging of the living transparent embryo. Here we describe a method to mount zebrafish embryos for long-term imaging and demonstrate how to automate the capture of time-lapse images using a confocal microscope. We also describe a method to create controlled, precise damage to individual...

We show the point-spread function of a confocal microscope with an increased detection aperture. The increase in aperture is accomplished by coherent collection of the light from the specimen with two opposing objective lenses, i.e., type-B 4Pi confocal microscopy. We demonstrate experimentally its feasibility for detecting scattered or fluorescently emitted light. The 4Pi confocal point-spread...

The purpose of this study was to determine whether in vivo images of oral mucosa obtained with a fiber optic confocal reflectance microscope could be used to differentiate normal and neoplastic tissues. We imaged 20 oral sites in eight patients undergoing surgery for squamous cell carcinoma. Normal and abnormal areas within the oral cavity were identified clinically, and real-time videos of eac...
