P-Nonylphenol (p-NP) as a non ionic surfactant is used in many industries such as pesticides, cosmetics, detergents and so on. Our previous study showed that the p-NP causes reduction in viability of the rats Bone Marrow Mesenchymal Stem cells (MSCs). The aim of the present study was to investigate the mechanism of the cell death due to p-NP exposure. The cells, after 3 passage were treated wit...
Acridine Orange is a cell-permeable green fluorophore that can be protonated and trapped in acidic vesicular organelles (AVOs). Its metachromatic shift to red fluorescence is concentration-dependent and, therefore, Acridine Orange fluoresces red in AVOs, such as autolysosomes. This makes Acridine Orange staining a quick, accessible and reliable method to assess the volume of AVOs, which increas...
Sorsby (I939) used intravenous Kiton green to produce permanent vital staining of the diseased human retina. Other workers have used vital staining dyes in experimental animals. Steinhausen and Loreth (I965) used Lissamin green in investigating the rat retina. Riehm and Podesta (I97 I) used both Patent blue (Disulphine blue) and Lissamin green in an examination of choroidal blood flow in an exp...
The impact of sperm DNA fragmentation on assisted reproductive technology (ART) successes, in terms of outcome, is now established. High levels of DNA strand breaks severely affect the probability of pregnancy. The importance of sperm nucleus condensation in early embryogenesis and, subsequently, on the quality of the conceptus is now emerging. In this article we have compared in situ analyses ...
INTRODUCTION: Micronuclei (MN) are microscopically visible round or oval cytoplasmic chromatin masses in the extra nuclear vicinity, originating from aberrant mitosis, chromatic fragments or whole chromosomes. The local factors like smoking exerts an influence on oral mucosa cells which are reflected as cellular changes resulting in the formation of MN. AIM: To evaluate and compare the MN index...
OBJECTIVE To determine whether Interferon-alpha-2b (IFN-α2b) can modulate the autophagic response in hepatocellular carcinoma cells. METHODS Hepatocellular carcinoma cells were treated with IFN-α2b. Autophagy was assessed by acridine orange staining, GFP-LC3 dotted assay, transmission electron microscopy and immunoblotting. RESULTS Acridine orange staining showed that IFN-α2b triggered the ...
BACKGROUND This study aimed to define the occurrence of different organisms causing vulvovaginitis; to evaluate different laboratory methods used for diagnosis of Trichomonas vaginalis (T. vaginalis); and to evaluate the direct score system and clue cell method compared with culture for diagnosis of bacterial and T. vaginalis vaginosis. METHODOLOGY Clinical and laboratory evaluations were per...
We have further characterized the sensitivity and specificity of SYPRO Orange protein gel stain and SYPRO Red protein gel stain with native and 2-dimensional polyacrylamide gels and for staining gels prior to Western blot analysis. We found that nucleic acids are not stained by the SYPRO protein gel stains, in contrast to results obtained with commonly used silver staining techniques. Bacterial...
Gram and acridine orange staining for diagnosis of septic arthritis in different patient populations
A new RNA-selective fluorescent dye integrated with a thiazole orange and a p-(methylthio)styryl moiety shows better nucleolus RNA staining and imaging performance in live cells than the commercial stains. It also exhibits excellent photostability, cell tolerance, and counterstain compatibility with 4',6-diamidino-2-phenylindole for specific RNA-DNA colocalization in bioassays.
