We applied two-photon polymerization to fabricate 3D synthetic niches arranged in complex patterns to study the effect of mechano-topological parameters on morphology, renewal and differentiation of rat mesenchymal stromal cells. Niches were formed in a photoresist with low auto-fluorescence, which enabled the clear visualization of the fluorescence emission of the markers used for biological d...

To study viability and activity of isolated common carp, Cyprinus carpio, ovarian follicular cells (granulosa and theca cells), 17- α -Hydroxy progesterone (17 α -OHP) and 17 β -Estradiol (E2) levels were estimated in the culture media of cultivated carp ovarian follicular cells, using radioimmunoassay (RIA). Oocytes were isolated from the ovaries of female carp. Interstitial tissue was manua...

Azadirachtin A in ppm quantity inhibits proliferation and monolayer formation of Spodoptera frugiperda (Sf9) insect cells in monolayer culture. The incubated cells demonstrate reduced rates of protein synthesis which finally leads to cell death. This growth inhibiting activity is compared with other botanicals such as ohchinin, salannin and volkensin. The high and specific activity of azadirach...

Transitional cell carcinoma of the urinary bladder is the second most common genitourinary malignancy in people in the United States. Cyclooxygenase-2 (COX-2) is overexpressed in bladder cancer. COX-2 inhibitors have had antitumor activity against bladder cancer, but the mechanisms of action are unclear. Clinically relevant concentrations of COX-2 inhibitors fail to inhibit proliferation in sta...

Culture supernatants of 30 enterotoxin-producing Bacillus cereus isolates produced a characteristic progressive destruction of McCoy cell monolayers. Enterotoxin-negative B. cereus and other group 1 Bacillus spp. caused no monolayer disruption. The McCoy cell tissue culture system appears to provide a rapid screening assay for detection of enterotoxin-producing B. cereus.

After seeding at subconfluent densities, human adrenocortical adenocarcinoma cells (SW-13) proliferate to form a typical epithelial monolayer upon the culture vessel substratum. Single cells of the confluent monolayer are spontaneously released into the medium, however, where they may remain isolated or reaggregate with other floating cells. In this study, we examined the growth patterns of the...

ex vivo expansion of human umbilical cord blood cells (hucbc) is explored by several investigators to enhance the repopulating potential of hucbc. the proliferation and expansion of human hematopoietic stem cells (hsc) in ex vivo culture was examined with the goal of generating a suitable clinical protocol for expanding hsc for patient transplantation. using primary human mesenchymal stem cells...

Erythrosin B and trypan blue are tested and compared for their effectiveness as vital exclusion stains for mammalian cells in monolayer culture. Both stains are supposed to mark cells that have lost membrane integrity. Fluorescein diacetate (FDA), an efficient vital inclusion stain, is used as a control, as it marks cells retaining membrane integrity. Erythrosin B and FDA are used as fluorescen...

Hepatocyte function on 3-D microfabricated polymer scaffolds realised with the pressure-activated microsyringe was tested under static and dynamic conditions. The dynamic cell culture was obtained using the multicompartment modular bioreactor system. Hepatocyte cell density, glucose consumption, and albumin secretion rate were measured daily over a week. Cells seeded on scaffolds showed an incr...

Triple negative breast cancer (TNBC) is a belligerent carcinoma that is unresponsive to targeted receptor therapies. Development of new treatment strategies would benefit from an expanded repertoire of in vitro cell culture systems, such as those that support tridimensional growth in the presence of hydrogel scaffolds. To this end, we established protocols for maintenance of the TNBC cell line ...
