To understand and characterize the pathogenic mechanisms of inflammatory bowel disease, dextran sulfate sodium (DSS) has been used to induce acute and chronic colitis in animal models by causing intestinal epithelium damage. The mechanism of action of DSS in producing this outcome is not well understood. In an effort to understand how DSS might impact epithelial cell metabolism, we studied the ...

It has been hypothesized that dysregulated host/microbial interactions play a pivotal role in the pathogenesis of inflammatory bowel disease. However, the exact mechanisms underlying the induction and perpetuation of the intestinal disorder are unclear. Recently, we unexpectedly discovered significantly upregulated gene expression of chitinase 3-like-1 in inflamed colon of the dextran sulfate s...

Dextran sulfate is semi-synthetic, polydisperse sulfated polysaccharide with important applications in clinical practice, in the manufacturing of plasma derived protein therapeutics and in biomedical research. The sensitive detection of dextran sulfate is relevant to preclinical and clinical drug development projects, the quality control of pharmaceutic formulations, and the process control in ...

Mice treated p.o. with 5% dextran sodium sulfate develop a mild to moderate colitis characterized by focal areas of inflammation and crypt abscesses. Immunohistological analysis of colons from dextran sodium sulfate-treated mice revealed an increased expression of intercellular adhesion molecule 1 (ICAM-1) and infiltration of lymphocyte function antigen 1-positive cells. A murine-specific antis...

The activity of intact urokinase (UK) and urokinases modified by soluble macromolecules (dextran and dextran sulfate sodium) in a mouse body was traced after injection of the 125I-labelled enzymes. The residual fraction of the enzyme in blood can be correlated with the time (t) as follows; Xb = Ae-at + Be-bt + Ce-ct Since a greater than b greater than c, the residual fraction of the enzyme in b...

BACKGROUND & AIMS In the intestines, Toll-like receptor 2 (TLR2) mediates immune responses to pathogens and regulates epithelial barrier function; polymorphisms in TLR2 have been associated with inflammatory bowel disease phenotype. We assessed the effects of TLR2 signaling on the enteric nervous system (ENS) in mice. METHODS TLR2 distribution and function in the ileal neuromuscular layer of ...

The interaction between dextran sulfate and low-density lipoproteins of the Sr 0-10 class in phosphate buffer of pH 7.4 and ionic strength 0.1 was studied by means of analytical ultracentrifugation. The sedimentation pattern of the dextran sulfate-lipoprotein mixture at high dextran sulfate/lipoprotein weight ratios showed a boundary indicative of a soluble complex with a high sedimentation rat...

This paper describes a simple method for purifying Coxiella burnetii cultivated in the chick embryo. The procedure is based upon the preliminary clarification of infected whole-egg suspensions by precipitating the low-density lipoproteins and other extraneous solids with dextran sulfate, calcium chloride, and sodium chloride solutions, and then concentrating the rickettsiae by centrifugation.

The effect of dextran sulfate on protein aggregation was investigated to provide the clues of its biochemical mechanism. The interaction between dextran sulfate and BSA varied with the pH values of the solution, which led to the different extent of aggregation prevention by dextran sulfate. Light scattering data with thermal scan showed that dextran sulfate suppressed BSA aggregation at pH 5.1 ...
