With this research we set out to develop a coumarin-based novel fluorescent probe NTR-AHC for the detection of biological thiols and nitroreductase (NTR). Probe was constructed by attaching NTR trigger moiety (p-nitrobenzyl) biothiol acceptor (maleic acid monoamide), core fluorophore AHC. Transfer ribonucleic acid (tRNA) fractions isolated from germinated and ungerminated spores of Botryodiplodia theobromae and Rhizopus stolonifer had acceptor activity for all 20 amino acids commonly found in protein, when tested with an enzyme fraction from germinated spores. NodA controls transfer of a fatty acid in the biosynthesis of lipochitin oligosaccharides by rhizobia. In an in vitro assay, we used de-N-acetylated chitin oligosaccharides substituted with an O-acetyl moiety as acyl acceptor substrates. We show that acyl-acyl carrier protein is used as a donor in NodA-directed fatty acid transfer.
