Concentrations of cycloheximide as low as 3 mug/ml inhibited incorporation of labeled orotic acid or uridine into RNA cytidylic acid of soybean (Glycine max) hypocotyl sections. Even lower concentrations of this well known protein synthesis inhibitor interfered with conversion of labeled cytidine into RNA uridylic acid. Both cycloheximide and puromycin inhibited absorption of (3)H-phenylalanine...

This report includes results demonstrating the existence of orotidine-5'-phosphate decarboxylase and orotidine-5'-phosphate pyrophosphorylase in plant leaves. The decarboxylase enzyme, purified 8 fold from leaves of etiolated pinto beans (Phaseolus vulgaris L.), had a pH optimum of 6.3. It was strongly inhibited by 6-azauridine-5'-phosphate; a concentration of 12 mum decreased the reaction rate...
The response of normal and regenerating liver to the hepatocarcinogen urethan was evaluated by means of an examination of the synthesis of free and membrane-bound ribosomal RNA. Partial hepatectomy results in a fiveto ninefold elevation of the incorporation of orotic acid-5-3H into free and membrane-bound ribosomal RNA 18 hr after operation; no noticeable changes occurred in the specific activi...
Orotic acid (OA) is an intermediate of pyrimidine nucleotide biosynthesis. Hereditary deficiencies in some enzymes associated with pyrimidine synthesis or the urea cycle induce OA accumulation, resulting in orotic aciduria. A link between patients with orotic aciduria and hypertension has been reported; however, the molecular mechanisms remain elusive. In this study, to elucidate the role of OA...
We demonstrated that a Klura3Δ, mutant of the yeast Kluyveromyces lactis is able to produce and secrete into the growth medium considerable amounts of orotic acid. Using yeast extract-peptone-glucose (YPD) based media we optimized production conditions in flask and bioreactor cultures. With cells grown in YPD 5% glucose medium, the best production in flask was obtained with a 1:12.5 ratio for f...
After a single injection of radioactive orotic acid and a "chase" of nonradioactive precursor, the specific activity of ribosomal RNA in rat liver decreases logarithmically at a rate corresponding to a half-life of about S days. The possible significance of this result is discussed with regard to control of protein synthesis.
The title compound, [Mn(C(5)H(2)N(2)O(4))(C(12)H(8)N(2))(H(2)O)(2)], was synthesized by the reaction of manganese(II) acetate and orotic acid in the presence of 1,10-phenanthroline. The crystal structure exhibits inter-molecular N-H⋯O and O-H⋯O hydrogen bonds . The Mn coordination environment consists of an N(3)O(3) donor set in an octa-hedral geometry.
