Increased lipogenesis is a hallmark of a wide variety of cancers and is under intense investigation as potential antineoplastic target. Although brisk lipogenesis is observed in the presence of exogenous lipids, evidence is mounting that these lipids may adversely affect the efficacy of inhibitors of lipogenic pathways. Therefore, to fully exploit the therapeutic potential of lipid synthesis in...

GH acts chronically on adipocytes to inhibit lipogenesis, antagonise insulin action and accentuate lipolysis. In addition, in some species, but not sheep, GH has a transient, acute, insulin-like effect, while for some preadipocyte-like cell-lines GH acts as a commitment factor. Studies on GH signalling in adipocytes have focused on these latter two effects and have implicated STAT proteins, MAP...

Lipogenesis is regulated by sterols and by insulin through the regulated expression and activation of the sterol regulatory element-binding proteins (SREBPs). A new study shows one way in which sterol and insulin regulation can be decoupled. In transgenic mice overexpressing a protein that regulates SREBP activation, lipogenesis is more sensitive to cholesterol and less sensitive to insulin.

AIM To determine the impact of paternal obesity, maternal obesity or the combination of two obese parents on markers of adult offspring metabolism, with a focus on body mass (BM), lipid and carbohydrate, components of lipogenesis and beta-oxidation in the liver, sex dimorphism in the offspring that received a SC diet during the postnatal period. MATERIALS AND METHODS Male and female C57BL/6 m...

The diurnal pattern of the activity of the pyruvate dehydrogenase complex (PDHC) was studied in the heart and liver of gold-thioglucose (GTG)-obese mice and age-matched controls. The diurnal pattern of lipogenesis was also measured in the liver. Both lean and obese mice had one main eating period, from 20:00 to 24:00 h. Eating produced no change in serum glucose of control mice but there was a ...

White-adipose-tissue lipogenesis showed a similar response to that of brown fat after glucose and insulin treatment in the three conditions, though the rates were severalfold lower in white adipose tissue. The differences in the basal lipogenesis in brown fat in the virgin, lactating and weaned states may be related to the plasma insulin concentration. Plasma insulin decreases during lactation ...

De novo lipogenesis and hypercaloric diets are thought to contribute to increased fat mass, particularly in abdominal fat depots. CB1 is highly expressed in adipose tissue, and CB1-mediated signalling is associated with stimulation of lipogenesis and diet-induced obesity, though its contribution to increasing fat deposition in adipose tissue is controversial. Lipogenesis is regulated by transcr...

Metabolic balance studies were carried out to determine the interrelationships of thyroid hormone-induced lipogenesis, lipolysis, and energy balance in the free-living rat. Intraperitoneal doses of 15 micrograms triiodothyronine (T3)/100 g body wt per d caused an increase in caloric intake from 26.5 +/- 1.7 (mean +/- SEM) kcal/100 g per d to 38.1 +/- 1.5 kcal/100 g per d. Food intake, however, ...
