Hepatitis C virus (HCV) envelope proteins E1 and E2 play an essential role in virus entry. However, the fusion mechanisms of HCV remain largely unclear, hampering the development of efficient fusion inhibitors. Here, we developed two cell-based membrane fusion models that allow for screening a peptide library covering the full-length E1 and E2 amino acid sequences. A peptide from the E2 stem do...

Amino-acid coevolution can be referred to mutational compensatory patterns preserving the function of a protein. Viral envelope glycoproteins, which mediate entry of enveloped viruses into their host cells, are shaped by coevolution signals that confer to viruses the plasticity to evade neutralizing antibodies without altering viral entry mechanisms. The functions and structures of the two enve...

Hepatitis C virus (HCV) is a causal agent of the chronic liver infection. To understand HCV morphogenesis, we studied the assembly of HCV structural proteins in insect cells. We constructed recombinant baculovirus expression vectors consisting of either HCV core alone, core-E1, or core-E1-E2. These structural proteins were expressed in insect cells and were examined to assemble into particles. ...

The fi rst prophylactic hepatitis C vaccine to reach human trials could enter phase II testing this year, a landmark for a fi eld that has faced many technical challenges since the virus was identifi ed in 1989. If this vaccine — or one of several other candidates — is successful, the public health impact could be substantial. Hepatitis C is a major cause of liver disease, including hepatocellu...

Hepatitis C virus (HCV) is a small enveloped virus whose genome is a RNA molecule encoding a polyprotein that is processed by cellular and viral proteases to produce the 3 structural proteins (the core protein C and the 2 envelope proteins E1 and E2) and the 6 nonstructural proteins (NS2, NS3, NS4A, NS4B, NS5A and NS5B). The HCV genome exhibits a significant genetic heterogeneity. HCV isolates ...

PURPOSE Chikungunya virus (CHIKV) causes endemic or epidemic outbreaks of CHIKV fever, which is a mosquitoe- transmitted viral disease in Africa, India, South-East Asia, and recently Southern Europe. Currently, serological diagnostic tests such as hemagglutination inhibition test (HI test), in-house IgM capture enzyme-linked immunosorbent assays (ELISA), and indirect immunofluorescence test wer...

We isolated East/Central/South African genotype chikungunya virus during the 2016 epidemic in Rio de Janeiro, Brazil. Genome sequencing revealed unique mutations in the nonstructural protein 4 (NSP4-A481D) and envelope protein 1 (E1-K211T). Moreover, all Brazil East/Central/South isolates shared the exclusive mutations E1-M407L and E2-A103T.
