Racemic and optically active 3-(2-deuteriophenyl)-2-(1-phenylpropan-2-yl)quinazoline-4-thiones were prepared. The nuclear magnetic resonance spectra clearly show that they exist as a 1:1 mixture of diastereomers due to the isotopic atropisomerism based on ortho-H/D discrimination (N–C axial chirality) chiral carbon.

A catalytic method has been developed for construction of indoline-2-thiones containing an all-carbon quaternary centre at the C-3 position. Successive treatment of α,β-unsaturated aldehydes bearing an isothiocyanato moiety with an in situ generated N-heterocyclic carbene and an appropriate heteroatomic nucleophile provided the 3,3-disubstituted indoline derivatives in moderate to good yields.
2-Dihydroquinazolin-4(1H)-thiones were prepared in up to excellent yields from 2-aminobenzothioamides and aldehydes. The reaction is carried out in water without the use of any catalyst or promoter. The sulfur-containing substrate can be obtained easily by thiation of the corresponding nitrile by solid sodium hydrosulfide.
