Native microorganisms present on grapes can influence final wine quality. Chambourcin is the most abundant hybrid grape grown in Pennsylvania and more resistant to cold temperatures fungal diseases compared Vitis vinifera. Here, non-Saccharomyces yeasts were isolated from spontaneously fermenting must three regional vineyards. Using cultured-based methods ITS sequencing, Hanseniaspora Pichia sp...

Although more than 3000 antibiotics have been reported from Actinomycetes/ higher fungi; biotechnological potential of yeasts and yeast-like fungi with respect to production of antimicrobial compounds has not been sufficiently investigated. We examined the antimicrobial activity of 11 strains of Aureobasidium pullulans (two new isolates and nine standard strains). All the strains of Aureobasidi...

A red-shifted, mutated form of the jelly-fish green fluorescent protein (GFP) under control of a TEF promoter was expressed at high levels in the filamentous fungus Aureobasidium pullulans. In the three transformants studied, all morphotypes of the fungus, including pigmented chlamydospores, expressed GFP and fluoresced brightly. Confocal microscopy showed that the intra-cellular distribution o...

The extracellular polygalacturonases produced by Aureobasidium pullulans isolated from waters of the Danube river were partially purified and characterized. The pH optima of polygalacturonases produced in the first phases of cultivation (48 h) and after 10 d as well as their optima of temperature, thermal stabilities, molecular masses, isoelectric points, action pattern and ability to cleave po...

In this study, two halophilic bacterial strains isolated from saline habitats in Argentina grew in the presence of gas oil. They were identified as Halomonas spp. and Nesterenkonia sp. by 16S ribosomal RNA sequencing. Chemotaxis towards gas oil was observed in Halomonas spp. by using swimming assays.

In this work, an ad hoc method to identify and quantify polyphenols from caper berries was developed on high-performance liquid chromatography/electrospray ionisation source/mass spectrometry (HPLC-ESI-MS). The method was applied during fermentation carried out with Lactobacillus pentosus OM13 (Trial S) and without starter (Trial C). A total of five polyphenols were identified. All samples cont...

Aureobasidium pullulans strain Y117 was transformed to hygromycin resistance using plasmid pDH33, which contains the bacterial hygromycin B phosphotransferase gene (hph) fused to promoter elements of the Aspergillus niger glucoamylase gene (glaA). Southern hybridizations of transformants revealed multiple, integrated copies of the vector. The glaA promoter was not induced by starch in A. pullul...

The production of pullulan from molasses and synthetic medium by Aureobasidium pullulans P56 in batch culture was investigated. Among the pretreatments of molasses used to increase polysaccharide production, sulfuric acid + activated carbon treatment gave better results with regard to polysaccharide concentration, polysaccharide yield and sugar utilization. A maximum polysaccharide concentratio...
