Previous studies of the conformations of peptides spanning the length of the alpha-spectrin SH3 domain suggested that SH3 domains lack independently folding substructures. Using a local structure prediction method based on the I-sites library of sequence-structure motifs, we identified a seven residue peptide in the src SH3 domain predicted to adopt a native-like structure, a type II beta-turn ...

Src Homology III (SHB) domains are small (55-?0 amino acids) receptor domains that are found in many signaling proteins and are involved in intracellular signaling events.t2 For example, the SH3 domain in the adaptor protein GRB-2 facilitates the coupling of growth factor receptors to an essential switch in growth-related signaling pathways-the guanine nucleotide binding protein Ras.3 Another e...

We report the complete genome sequence of Exiguobacterium sp. strain MH3, isolated from the rhizosphere of duckweed. The genome assembly is 3.16 Mb, with a G+C content of 47.24%, and it may provide useful information about plant-microbe interactions and the genetic basis for the tolerance of the strain to various environmental stresses.

Domain insertions and deletions lead to variations in the domain architectures of the proteins from their common ancestor. In this work, we investigated four groups of the RhoGEF-containing proteins from different organisms with domain architectures RhoGEF-PH-SH3, SH3-RhoGEF-PH, RhoGEF-PH, and SH3-RhoGEF defined in the Pfam database. The phylogenetic trees were constructed using each individual...

We report here the NMR derived structure of the binary complex formed by the Itk SH3 and SH2 domains. The interaction is independent of both a phosphotyrosine motif and a proline-rich sequence; the classical targets of the SH2 and SH3 domain, respectively. The Itk SH3/SH2 structure reveals the molecular details of this non-classical interaction and provides a clear picture for how the previousl...

RhoGEFs are central controllers of small G-proteins in cells and are regulated by several mechanisms. There are at least 22 human RhoGEFs that contain SH3 domains, raising the possibility that, like several other enzymes, SH3 domains control the enzymatic activity of guanine nucleotide exchange factor (GEF) domains through intra- and/or intermolecular interactions. The structure of the N-termin...

Exiguobacterium sp. KKBO11, isolated near a wastewater treatment plant in Houston, Texas, USA, possesses a large number of genes involved in stress response and transport critical to survival in adverse environmental conditions. An unusually high copy number of RNA genes also possibly contributes to this microorganism's versatility by promoting nutrient uptake.

High-quality draft genome sequences were determined for 10 Exiguobacterium strains in order to provide insight into their evolutionary strategies for speciation and environmental adaptation. The selected genomes include psychrotrophic and thermophilic species from a range of habitats, which will allow for a comparison of metabolic pathways and stress response genes.

A number of peroxisome-associated proteins have been described that are involved in the import of proteins into peroxisomes, among which is the receptor for peroxisomal targeting signal 1 (PTS1) proteins Pex5p, the integral membrane protein Pex13p, which contains an Src homology 3 (SH3) domain, and the peripheral membrane protein Pex14p. In the yeast Saccharomyces cerevisiae, both Pex5p and Pex...
