In vivo electroporation of tumours shows disruption of blood flow and creates a vascular effect with an initial rapid and transient vasoconstriction phase and a much longer lasting phase with changed microvascular endothelium. These changes are not well understood but are presumed to involve the cytoskeleton. The paper presents for the first time differential in vitro effects describing cytoske...
Local management of soft tissue sarcoma in humans generally involves multi-modality approaches whose cornerstones are surgery combined with radiation therapy. The usual radiation protocols are based on preoperative, intraoperative, or postoperative external beam treatment or adjuvant brachytherapy. The aim of these strategies is to maximize tumor control while minimizing side effects, especiall...
Electrochemotherapy increases the permeability of tumours to drugs by electric voltages applied locally. Its value in tumours of the head and neck is unknown. We retrospectively reviewed a 2-centre database, and found 39 patients with squamous cell carcinoma (SCC) of the oral cavity or oropharynx (n=12) or non-melanoma skin tumours (n=27) who had been treated with bleomycin electrochemotherapy ...
BACKGROUND The aim of this study was to investigate the antitumor effectiveness of electrochemotherapy with cisplatin combined with suboptimal radiotherapy doses. Tumor radiosensitization was evaluated on large invasive ductal carcinoma tumors in Balb/C mice. MATERIALS AND METHODS Tumors of an average volume of 630 mm(3) were treated with cisplatin, electric pulses, radiotherapy, electrochemo...
Electrochemotherapy is an effective antitumor treatment employing locally applied high voltage electric pulses delivered through conductive electrodes to the tumor in combination with chemotherapeutic drugs. The efficiency of electrochemotherapy strongly depends on the local electric field distribution inside the target tissue. For successful therapy the entire target tissue has to be exposed t...
The treatment of soft tissue tumors needs the coordinated adoption of surgery with radiation therapy and eventually, chemotherapy. The radiation therapy (delivered with a linear accelerator) can be preoperative, intraoperative, or postoperative. In selected patients adjuvant brachytherapy can be adopted. The goal of these associations is to achieve tumor control while maximally preserving the n...
Electrochemotherapy is now in development for treatment of deep-seated tumors, like in bones and internal organs, such as liver. The technology is available with a newly developed electric pulse generator and long needle electrodes; however the procedures for the treatment are not standardized yet. In order to describe the treatment procedure, including treatment planning, within the ongoing cl...
Muscle contractions present the main source of unpleasant sensations for patients undergoing electrochemotherapy. The contractions are a consequence of high voltage pulse delivery. Relatively low repetition frequency of these pulses (1 Hz) results in separate muscle contractions associated with each single pulse that is delivered. It would be possible to reduce the number of unpleasant sensatio...
We demonstrate feasibility and safety of Electrochemotherapy for treatment of a prostate cancer (PCa) with infiltration of the urethral sphincter. The patient remained continent and potent, toxicity was low, and 6 months of follow-up showed no cancer activity. We conclude that Electrochemotherapy should be further evaluated as treatment strategy for locally advanced PCa.
