Several congenital disorders may cause neutropenia. The recent advances in the field of molecular biology have facilitated our knowledge concerning pathophysiological mechanisms leading to these disorders. The molecular basis of disorders with defective myelopoiesis (severe congenital neutropenia and cyclic neutropenia) and disorders due to defective RNA processing (Shwachman Diamond syndrome a...
Cyclic neutropenia is a rare haematologic disorder. This case report presents a 6-year-old girl who was referred in order to treat gingival recession, and was finally diagnosed as having cyclic neutropenia based on an oral examination, complete blood counts and a sternal puncture bone marrow test. The patient received systematic granulocyte colony-stimulating factor therapy, full-mouth scaling ...
A 78-year-old woman diagnosed with cyclic neutropenia 5 years previously had been treated with recombinant granulocyte-colony stimulating factor (G-CSF). She developed fever, tenderness and distension of temporal arteries after the treatment with G-CSF. Magnetic resonance imaging and ultrasonography revealed wall thickening of the temporal arteries. She was therefore diagnosed with giant cell a...
Mutations in ELA2 encoding the neutrophil granule protease, neutrophil elastase (NE), are the major cause of the 2 main forms of hereditary neutropenia, cyclic neutropenia and severe congenital neutropenia (SCN). Genetic evaluation of other forms of neutropenia in humans and model organisms has helped to illuminate the role of NE. A canine form of cyclic neutropenia corresponds to human Hermans...
A child with cyclical neutropenia was observed before and during treatment with levamisole. Marked clinical improvement occurred while on levamisole. Deterioration occurred after stopping treatment. The improvement was not associated with changes in the neutrophil or monocyte counts, nor did tests of phagocytic cell function convincingly demonstrate improvement while the child was on treatment.
BACKGROUND AND OBJECTIVES The two main complications of severe chronic neutropenia are fatal sepsis and myelodysplasia/acute leukemia (MDS/AL). Granulocyte colony-stimulating factor (G-CSF) therapy has significantly reduced the frequency and severity of infections, but its possible influence on the risk of malignancy is not known. DESIGN AND METHODS The French Severe Chronic Neutropenia (SCN)...
