Toxoplasma gondii proliferates and organizes within a parasitophorous vacuole in rosettes around a residual body and is surrounded by a membranous nanotubular network whose function remains unclear. Here, we characterized structure and function of the residual body in intracellular tachyzoites of the RH strain. Our data showed the residual body as a body limited by a membrane formed during prol...
BACKGROUND Toxoplasma gondii is one of the most common causes of latent infections in humans worldwide. Detecting anti-Toxoplasma antibodies in serum using serological tests is a common method to diagnose toxoplasmosis. OBJECTIVES In the present study, an indigenous ELISA kit was prepared using tachyzoites from the RH strain of T. gondii, and its sensitivity and specificity were compared with...
To evaluate the usefulness of the Korean Isolate-1 (KI-1) antigen for serodiagnosis of toxoplasmosis, antigen profiles of KI-1 tachyzoites were analyzed in comparison with RH tachyzoites by SDS-PAGE and immunoblotting. ELISA was performed on latex agglutination (LA)-positive and negative serum samples using KI-1 and RH antigens. Immunoblotting of the KI-1 antigen showed multiple antigen bands w...
background: serological diagnosis of toxoplasma gondii infection using crude antigens may not be more accurate. to increase the diagnostic potency of anti-gens, isolation of their immunogenic fractions could be useful. the current re-search adopted to obtain an affinity isolated fraction from rh strain using cnbr sepharose 4b column coupled with infected mice sera helping in detec-tion of igm a...
We studied the pathology of acute toxoplasmosis in experimental mice inoculated with RH strain tachyzoites of Toxoplasma gondii. All died from severe disseminated toxoplasmosis involving the liver, spleen and pancreas. Pathological features of acute toxoplasmosis in susceptible mice could be regarded as an excellent model for acute reactivation of Toxoplasma in the immunosuppressed host.
Objectives: Toxoplasma gondii is an obligate intracellular parasite that infects a broad range of warmblooded animals including human. Tachyzoites of T.gondii invade the host cell, replicate and finally lead to the lysis of the cell. T. gondii is associated with congenital infection and it can cause encephalitis, or systemic infection in immunocompromised patients. It is important to know wheth...
Immunization with DNA can induce humoral and cell-mediated immune responses, both of which are important in conferring immunity to Toxoplasma gondii. The efficacy of genetic vaccination with a cDNA encoding the T. gondii SAG1 (P30) surface antigen was evaluated. Sera of immunized mice showed recognition of T. gondii tachyzoites by immunofluorescence and exhibited high titers of antibody to SAG1...
Study of resistance to Toxoplasmosis using Tachizoites surface antigens of Toxoplasma gondii in vivo
Taoxoplasma gondii is an important food-borne parasite transmitted primarily from animals to humans through meat consumption, mainly pork and lamb, as well as through oocysts shed by cats. Infection in humans can cause severe fetal malformations, ocular complications or encephalitis. Toxoplasmosis infection during pregnancy, especially in sheep, often results in abortion, representing considera...
AIM To determine the usefulness of prednisolone in increasing the number of Toxoplasma (T.) gondii tachyzoites and bradyzoites in mice. MATERIALS AND METHODS The mice were water-fasted prior to being immunosuppressed with oral inoculation of prednisolone. Tachyzoites of 7T gondii RH strain were inoculated into mice and the number of the parasites in the intraperitoneal fluids was then determi...
Genetic typing methods of T. gondii strains have been extensively perfected in recent years. From a technical point of view, many tools usable for genetic studied on single-copy loci have been used: RFLP, PCR-RFLP, sequencing, RAPD-PCR and isoenzyme analysis. We described the cloning and sequence analysis of the gene which encodes the major surface antigen (SAG1 or P30) of T. gondii. SAG1 is ...
