Haemophilus ducreyi, the etiological agent of chancroid, has a strict requirement for heme, which it acquires from its only natural host, humans. Previously, we showed that a vaccine preparation containing the native hemoglobin receptor HgbA purified from H. ducreyi class I strain 35000HP (nHgbAI) and administered with Freund's adjuvant provided complete protection against a homologous challeng...

We report evidence of dense, ordered nanodomains in single-component fluid lipid bilayers. Our atomic-scale molecular dynamics simulations suggest that the area available to a lipid acyl chain exhibits large fluctuations, resulting in denser and sparser domains. The sizes of the dense domains can be up to approximately 10 nm, and their lifetimes are of the order of approximately 10 ns. In addit...

The formation and dynamics of spatially extended compositional domains in multicomponent lipid membranes both in vivo and in vitro lie at the heart of many important biological and biophysical phenomena. While the thermodynamic basis for domain formation has been explored extensively in the past, the roles of membrane and exterior fluid hydrodynamics on domain formation kinetics have received l...

An immunosensing device, comprising a lipid membrane incorporating ion channels tethered to the surface of a gold electrode, has been reported [Cornell, Braach-Maksvytis, King, Osman, Raguse, Wieczorek and Pace (1997) Nature (London) 387, 580-583]. The present article describes key steps in the assembly of the device and provides further evidence for its proposed sensing mechanism.

A multilayered-substrate model is proposed for surface plasmon resonance (SPR)-based measurement of the thickness of a water layer sandwiched between a lipid bilayer and an underlying support. To calculate sensitivity, a 473 nm-wavelength excitation source and a silver layer are used for the SPR sensor. It is theoretically shown that the multilayered substrate design achieves sufficient sensiti...

The competition between chain entropy and bending rigidity in compact polymers can be addressed within a lattice model introduced by Flory in 1956 [Proc. R. Soc. London A 234, 60 (1956)]]. It exhibits a transition between an entropy dominated disordered phase and an energetically favored crystalline phase. The nature of this order-disorder transition has been debated ever since the introduction...

In this report we briefly describe an immunoglobulin G3 monoclonal antibody, 2G6/1H11, which binds purified lipid A from Salmonella minnesota and a lipid A precursor molecule derived from Salmonella typhimurium. 2G6/1H11 does not bind well to purified whole S. typhimurium lipopolysaccharide (LPS), S. minnesota LPS, LPS preparations from a series of S. minnesota rough mutants, or intact S. typhi...

We present a simple, and physically motivated, coarse-grained model of a lipid bilayer, suited for micron scale computer simulations. Each approximately 25 nm(2) patch of bilayer is represented by a spherical particle. Mimicking forces of hydrophobic association, multiparticle interactions suppress the exposure of each sphere's equator to its implicit solvent surroundings. The requirement of hi...

As a surrogate of live cells, proteo-lipobeads are presented, encapsulating functional membrane proteins in a strict orientation into a lipid bilayer. Assays can be performed just as on live cells, for example using fluorescence measurements. As a proof of concept, we have demonstrated proton transport through cytochrome c oxidase.
