Internalin A is an essential virulence gene involved in the uptake of the foodborne pathogen Listeria monocytogenes into host cells. It is intact in clinical strains and often truncated due to Premature Stop Codons (PMSCs) in isolates from processed foods and processing facilities. Less information is known about environmental isolates. We sequenced the inlA alleles and did Multi Locus Variable...
Animal models are generalized linear mixed model (GLMM) used in evolutionary biology and animal breeding to identify the genetic part of traits. Integrated Nested Laplace Approximation (INLA) is a methodology for making fast non-sampling based Bayesian inference for hierarchical Gaussian Markov models. In this paper we demonstrate that the INLA methodology can be used for many versions of Bayes...
The role of sigma(B) in Listeria monocytogenes infection of human intestinal epithelial cells was investigated. Invasion defects associated with loss of sigma(B) paralleled those of a DeltainlA strain independently of the sigma(B)-dependent P2(prfA) promoter. Concomitantly, amounts of inlA transcript and InlA protein were significantly decreased in the DeltasigB strain.
In this report we apply Integrated Nested Laplace approximation (INLA) to a series of multivariate stochastic volatility models. These are a useful construct in financial time series analysis and can be formulated as latent Gaussian Markov Random Field (GMRF) models. This popular class of models is characterised by a GMRF as the second stage of the hierarchical structure and a vector of hyperpa...
Listeria monocytogenes mutants with deletions in aroA, aroB, or aroE exhibited strong posttranscriptional upregulation of internalin A (InlA) and InlB synthesis, which resulted in a more-than-10-fold increase in InlA-mediated internalization by epithelial Caco-2 cells and a 4-fold increase in InlB-mediated internalization by microvascular endothelial cells (human brain microvascular endothelial...
Internalin A (InlA), a protein required for Listeria monocytogenes virulence, is encoded by the inlA gene, which is only found in pathogenic strains of this genus. One of the best ways to detect and confirm the pathogenicity of the strain is the detection of one of the virulence factors produced by the microorganism. This paper focuses on the design of an electrochemical genosensor used to dete...
Eighty Listeria monocytogenes isolates were obtained from Chinese retail ready-to-eat (RTE) food and were previously characterized with serotyping and antibiotic susceptibility tests. The aim of this study was to characterize the subtype and virulence potential of these L. monocytogenes isolates by multilocus sequence typing (MLST), virulence-associate genes, epidemic clones (ECs), and sequence...
A panel of hybridomas was produced using intact Listeria monocytogenes serotype 1/2a cells as the immunogen. An IgG2a monoclonal antibody (mAb) 'mAb2B3' was isolated that reacted with L. monocytogenes but not with a representative panel of related Listeria spp. and non-Listeria spp. Binding activity was greatest against L. monocytogenes serotype 1/2a and was significantly enhanced when cells we...
Integrated Nested Laplace Approximations (INLA) has been a successful approximate Bayesian inference framework since its proposal by Rue et al. (2009). The increased computational efficiency and accuracy when compared with sampling-based methods for like MCMC methods, are some contributors to success. Ongoing research in the INLA methodology implementation thereof R package R-INLA, ensures cont...
Animals are important reservoir of Listeria monocytogenes, a pathogen causing serious infections in both humans and livestock. However, data on invasiveness of L. monocytogenes strains of animal origin is very scarce. Ability of 18 L. monocytogenes strains of animal origin to invade HT-29 cells was investigated. Plaque forming assay was used to assess invasiveness and ability of the pathogen to...
