In situ hybridization (ISH) is a method that uses labeled complementary single strand DNA or RNA to localize specific DNA or RNA sequences in an intact cell or in a fixed tissue section. The main steps of ISH consist of: probe selection, tissue or sample preparation, pre-hybridization treatment, hybridization and washing, detection and control procedure. Probe selection is one of the important ...

A fluorescent probe (N-(4-methyl-2-oxo-2H-chromen-7-yl)-2,4-dinitrobenzenesulfonamide), which exhibits high selectivity to glutathione and cysteine among amino acids including sulphur-containing methionine and metal ions, was synthesized. The experiments demonstrate that the fluorescent probe is a reliable and specific probe for glutathione and cysteine in living cells.

Subcellular microenvironment plays a crucial role in maintaining cell homeostasis being hydration an essential parameter to monitor the variations of lipid organization biological membranes. Fluorescent microenvironment-sensitive probes are stimuli-responsive molecules and able change their fluorescence profile according properties (polarity, hydration, microviscosity tension). Here we present ...

A new ‘off-on’ type Al-selective fluorescent probe derived from benzoyl hydrazine has been synthesized and characterized. This probe shows high selectivity towards Al compared to other common metal ions, and complexing with Al triggers a new bond at 450 nm. With optimized conditions, the probe gives a linear response of 8.9×10–9.0 ×10M with a detection limit of 2.9×10 M Al in ethanol. Key-Words...

There is a pressing need in neuroscience for genetically-encoded, fluorescent voltage probes that can be targeted to specific neurons and circuits to allow study of neural activity using fluorescent imaging. We created 90 constructs in which the voltage sensing portion (S1-S4) of Ciona intestinalis voltage sensitive phosphatase (CiVSP) was fused to circularly permuted eGFP. This led to Electric...
