Herein, we firstly demonstrate the design and the proof-of-concept use of a capillary-driven surface-enhanced Raman scattering (SERS)-based microfluidic chip for abrin detection. The micropillar array substrate was etched and coated with a gold film by microelectromechanical systems (MEMS) process to integrate into a lateral flow test strip. The detection of abrin solutions of various concentra...

teins on the tumor cells was shown to be specific, pH de pendent, and concentration dependent. Native ricin, abrin, D-galactose, and its sterically related saccharides inhibit this specific binding, while concanavalin A, bovine serum albumin, heat-denatured abrin or ricin, and other sac charides do not. In the presence of D-galactose, the inhibi tion of protein biosynthesis by ricin does not oc...

abrin known as ribosome inactivating protein’s (rip) it is a high cytotoxic plant protein. high lethality and low cost and easy access to plant and seeds lead to this toxin used in crimes and terrorist acts. because obtaining purified toxins requires advanced laboratory equipment and complex procedures, so, it appears that the perpetrators of such crimes use crude extracts. as a result, remaini...

Seeds of Gunja (Abrus precatorius Linn) are reported to be poisonous due to the presence of a toxic protein 'abrin and an alkaloid hypaphorine. Te seeds after subjecting to the process of shodhana, find use in a number of Ayurvedic preparations. HPLC studies revealed that the process of shodhana resulted in depletion of more toxic alkaloid hypaphorine and protein abrin.

Abrin, one of the most highly potent toxins in the world, is derived from the plant, Abrus precatorius. Because of its high toxicity, it poses potential bioterror risks. Therefore, a need exists for new reagents and technologies that would be able to rapidly detect abrin contamination as well as lead to new therapeutics. We report here a group of abrin-specific monoclonal antibodies (mAbs) that...

A non-toxic dose of abrin, (1.25 microg/kg body wt) consecutively for five days in normal mice stimulated specific humoral responses. A noticeable increase was observed in total leucocyte count, lymphocytosis, weights of spleen and thymus, circulating antibody titre, antibody forming cells, bone marrow cellularity and alpha-esterase positive bone marrow cells. The results suggest that abrin can...

A panel of immunotoxins was constructed by chemically attaching the ribosome-inactivating proteins abrin A chain, ricin A chain, gelonin, and momordin to the monoclonal mouse IgG2a antibody Fib75 by means of a disulfide linkage. All the immunotoxins were toxic in tissue culture to the I.I human bladder carcinoma cell line expressing the antigen recog nized by Fib75, inhibiting the incorporation...

Abrin belongs to the type II family of ribosome-inactivating proteins comprising a galactose-binding B chain coupled with a toxic A chain through a single disulphide linkage. Apart from its RNA-N-glycosidase activity, another role that has been recently ascribed to abrin was the induction of apoptosis. Studies were undertaken to determine the kinetics of these two activities. In the present stu...

The protein toxin abrin, which possesses an N-glycosylase activity toward eukaryotic 28 S rRNA, may have a potential in the deliberate eradication of certain cells. Here we report, by polymerase chain reaction technique, the isolation of genomic DNA sequences encoding two distinct abrin A-chains. A third sequence which encoded a part of a third type of A-chain was also isolated. The deduced ami...
