Recent studies have identified a class of small non-coding RNA molecules, named microRNA (miRNA), that is dysregulated in malignant brain glioblastoma. Substantial data have indicated that miRNA-16 (miR-16) plays a significant role in tumors of various origins. This miRNA has been linked to various aspects of carcinogenesis, including cell apoptosis and migration. However, the molecular functio...

Inflammation is the body's normal self-protection mechanism to eliminate pathogens and resist pathogen invasion. The excessive inflammatory response may lead to inflammatory lesions. The mechanisms accounting for inflammation remain hazy. miRNAs have been proposed to have crucial effects on inflammation. In the present study, we reported that lipopolysaccharide (LPS)-stimulation increased the e...

The role of specific members of the NF-κB family of transcription factors in CD8 T-cell selection and development is largely unknown. Here, we show that mice lacking NF-κB1 develop a unique population of conventional CD8 single-positive (SP) thymocytes with memory T cell-like properties that populate peripheral immune organs. Development of this memory-like population is not due to PLZF(+) thym...

Purpose: To investigate the effect of microRNA-16 (miR-16) on glioma cell migration and invasiveness, mechanism involved. Methods: MicroRNA-16 mimic or inhibitor was transfected into human (SHG44) cells. Cell migration, invasiveness morphology were determined using scratch test, Transwell invasion assay, immunohistochemical staining, respectively. Expressions bcl-2, MMP-9 MMP-2, NF-κB1 proteins ...

NF-κB is a key transcriptional regulator involved in inflammation and cell proliferation, survival, and transformation. Several key steps in its activation are mediated by the ubiquitin (Ub) system. One uncharacterized step is limited proteasomal processing of the NF-κB1 precursor p105 to the p50 active subunit. Here, we identify KPC1 as the Ub ligase (E3) that binds to the ankyrin repeats doma...

Numerically, five divisions of transcription factor family have been classified. These are tagged as p65 (RelA), RelB, c-Rel, NF-κB1 and NF-κB2. In dissimilarity to the other family divisions, NF-κB1 and NF-κB2 are produced in inactive forms (p105 and p100) and are enzymatically broken down to the active forms (p50 and p52), respectively [8]. All the five divisions of this protein family tree f...

BACKGROUND Previous investigations and meta-analyses on the effect of glucocorticoids on mortality in septic shock revealed mixed results. This heterogeneity might be evoked by genetic variations. Such candidate is a promoter polymorphism (-94ins/delATTG) of the gene encoding the ubiquitous transcription-factor nuclear-factor-κB (NF-κB) which binds to recognition elements in the promoter of sev...

nf-κb pathway is a link between inflammation and cancer and is involved in cellular responses to different stimuli. gastrointestinal lumen is exposed to many inflammatory agents such as foods, free radicals and bacterial or viral antigens. the aim of the present study was to evaluate the possible role of nf-κb1in gastric adenocarcinoma. to detect the relative level of nf-κb1transcript, total rn...

NF-κB proteins play a complex role in modulating carcinogenesis following DNA damage. Previous work identified p50/NF-κB1 as a necessary factor in the cytotoxic response to alkylation damage. Recently, these findings were extended to demonstrate that in the setting of alkylation damage, this NF-κB subunit acts as a haploinsufficient tumor suppressor that prevents hematologic malignancy formation.

Although NF-κB1 p50/p105 has critical roles in immunity, the mechanism by which NF-κB1 regulates inflammatory responses is unclear. Here, we analyzed the gene expression profile of lipopolysaccharide (LPS)-stimulated Nfkb1−/− macrophages that lack both p50 and p105. Deficiency of p50/p105 selectively increased the expression of interferon (IFN)-responsive genes, which correlated with increased ...
