UNLABELLED Coxiella burnetii is an intracellular pathogen that replicates within a lysosome-like vacuole. A Dot/Icm type IVB secretion system is used by C. burnetii to translocate effector proteins into the host cytosol that likely modulate host factor function. To identify host determinants required for C. burnetii intracellular growth, a genome-wide screen was performed using gene silencing b...

Background: Pah1p is a phosphatidic acid phosphatase that generates diacylglycerol. Results: Deletion of PAH1 alters the sorting of fusion factors to the vacuole and inhibits fusion. Conclusion: Conversion of phosphatidic acid to diacylglycerol is integral to vacuole homeostasis. Significance: This is the first report that links a Lipin1 homologue to the recruitment and activation of a Rab GTPa...

The ultrastructural localization of peroxidase was studied in guinea pig eosinophils which had phagocytized zymosan or Escherichia coli. After phagocytosis, the membrane of the granule was joined to the membrane of the phagocytic vacuole, and the enzyme, which is ordinarily restricted to the matrix of the granule, was seen in the vacuole surrounding the ingested particle.

The cells of higher plants, in contrast with those of other organisms, contain relatively large amounts of organic acids. Krebs & Lowenstein (1960) calculated that the concentrations of individual organic acids in Escherichia coli are about 0-01 mM, in liver or kidney cells 0 1 mm, and in yeast cells 1 mm. By using the same type of calculation it can be estimated that many plant tissues contain...

Fluorescein isothiocyanate-conjugated dextran (FITC-dextran) is internalized by endocytosis into the lysosome-like vacuoles of Saccharomyces cerevisiae (Makarow, M., 1985, EMBO (Eur. Mol. Biol. Organ.) J. 4:1861-1866). Here we show that under energy depletion conditions FITC-dextran accumulated in a cytoplasmic compartment, from which it could be chased to the vacuole when the energy block was ...
