What is blotting / hybridization? Blotting of nucleic acid is the central technique for hybridization studies. Double stranded DNA will denature or separate at high temperatures into single strands. When the temperature is lowered, the two strands will anneal because of the base pairing interaction between the complementary strands. If nucleotide sequences are identical or similar, complementar...

A consideration of the earlier investigations of the nucleic acids would lead us to the conclusion that these substances yield among their hydrolytic products four purin derivatives, namely, xanthin, hypoxanthin, guanin, and adenin. Of these four bases hypoxanthin was the first discovered in this connection and was for a long time looked upon as the most constant in its occurrence, and, therefo...

Fluorescent nucleic acid base analogues and the corresponding nucleosides that reveal excited-state proton transfer, including phototautomerism, are briefly characterized, and their potential applications as fluorescent probes discussed. Among the most promising systems are formycins and some 8-azapurines as well as the corresponding nucleosides, like 8-azaxanthosine. Other, nonfluorescent syst...

Graphene oxide quenches fluorescence corresponding to only a mismatched target due to selective denaturing of the thermo-unstable duplex composed of probe peptide nucleic acid and single base mismatched target RNA and thus, the fluorescence signal only from perfectly matched target RNA is measured.

We have combined an allosteric molecular beacon for target recognition and guanine-rich DNAzyme for signal amplification to develop a new platform for visual detection of nucleic acids with single-base mismatch detection capability. The fully DNA-structured platform can undergo color change in response to target DNA/RNA, which enables sensitive and selective visual detection in biological samples.

Nucleic acids include substantial information in their base sequence and their hybridization-complexation motifs. Recent research efforts attempt to utilize this biomolecular information to develop DNA nanostructures exhibiting machine-like functions. DNA nano-assemblies revealing tweezers, motor, and walker activities exemplify a few such machines. The DNA-based machines provide new components...

Ultra-high molecular weight DNA/polymer hybrid materials were prepared employing molecular biology techniques. Nucleic acid restriction and ligation enzymes were used to generate linear DNA di- and triblock copolymers that contain up to thousands of base pairs in the DNA segments.

In order to better understand the adsorption mechanism of nucleic acids at electrode surface, MAC Mode atomic force microscopy (MAC Mode AFM) was used to investigate the adsorption of a 10-base synthetic oligodeoxynucleotide (ODN) with the sequence 5′-CTTTTTCTTT-3′ containing o a T e a i i ©

Recombinant human immunodeficiency virus type 1 (HIV-1) Gag protein can assemble into virus-like particles (VLPs) in suitable buffer conditions with nucleic acid. We have explored the role of nucleic acid in this assembly process. HIV-1 nucleocapsid protein, a domain of Gag, can bind to oligodeoxynucleotides with the sequence d(TG)(n) with more salt resistance than to d(A)(n) oligonucleotides. ...
