An asymmetric route for the synthesis of highly functionalized 2,3-epoxy-syn-1,4cyclohexane diol derivatives present in some polyketide natural products has been developed. The key step involves RCM of an appropriately constructed 1,7-dienol derived from D-mannitol to cyclohexane1,4-diol followed by its stereoselective epoxidation.
[reaction: see text] The callipeltoside A chlorocyclopropyl-containing dienyne side chain has been synthesized in nine steps and 33% overall yield from commercially available 1,2,5,6-O-dicyclohexylidene-D-mannitol. The key steps in the synthesis are a highly diastereoselective cyclopropanation of a vinyl chloride allylic ether and a Suzuki cross-coupling to complete the carbon framework.
[structure: see text] A new chiral ferrocenyl diphosphine ligand 3 was synthesized from readily available D-mannitol. Rh-complex with this ligand showed high enantioselectivity and reactivity in the asymmetric hydrogenation of dehydroamino acid derivatives and itaconic acid derivatives.
