A 419-bp region of the flagellin gene sequence of Borrelia burgdorferi was used as a target for the polymerase chain reaction. With a nonradioactively labeled gene-specific probe, sensitivity to as few as 1 to 10 spirochetes was observed. The targeted gene fragment was conserved in the American and European strains of B. burgdorferi tested and among several other pathogenic borreliae.

We have characterized a Borrelia burgdorferi clone (CA-11 2A) that does not synthesize the major outer surface proteins, OspA and OspB. While the osp operon is intact and capable of expression, no mRNA transcript is detectable in this clone. These results suggest that osp operon expression in the B. burgdorferi clone can be regulated at the level of transcription.

We evaluated the new MPM medium for the growth of Borrelia burgdorferi. All 18 blood samples from 17 patients with Lyme disease were negative. Growth studies showed that by day 4, most organisms in MPM were not viable. Our results reinforce the use of BSK medium as the primary choice for growing B. burgdorferi.

We previously demonstrated that Borrelia burgdorferi requires OspC to colonize a mammalian host. To delineate this requirement, we analyzed the clearance of ospC mutant spirochetes and found that they were eliminated within 48 h. We conclude that B. burgdorferi uses OspC to resist innate host defenses immediately after transmission.
