We investigated HLA DRB1 correlations with chronic lymphocytic leukemia (B-CLL) outcome in 90 patients. Neither of the alleles was associated with B-CLL clinical characteristics or mortality. HLA DRB1*01 and HLA DRB1*02-null were associated with shorter overall survival (p=0.007, p=0.002). Our results suggest that HLA-restricted adaptive immunity influences CLL outcome.

The responsiveness and diversity of peripheral B-cell repertoire decreases with age, possibly because of B-cell clonal expansions, as suggested by the incidence of serum monoclonal immunoglobulins and of monoclonal chronic lymphocytic leukemia (CLL)-like B lymphocytes in clinically silent adults. We phenotyped peripheral blood cells from 500 healthy subjects older than 65 years with no history ...

B-cell chronic lymphocytic leukemia (CLL) is characterized by differential BCR signaling and autoimmune complications. Complement modulates B-cell function via C3d and CD21 cross-linked to the B-cell receptor (BCR). We hypothesized that CD21 contributes to BCR signaling and participates in the autoimmunity associated with CLL. We analyzed CD21 expression on 106 CLL patient samples and matched s...

Among the hematologic malignancies, the genetic lesion in chronic myelogenous leukemia (CML) resulting in Bcr-Abl tyrosine kinase has become a hallmark target for therapeutic intervention. Inhibition of Bcr-Abl tyrosine kinase is a poster child for kinasetargeted therapeutics. Unlike CML, chronic lymphocytic leukemia (CLL) does not have a genetic lesion associated with pathophysiology or biolog...

B-chronic lymphocytic leukemias (B-CLL) represent expanded clones of resting B lymphocytes that mostly express surface IgM (sIgM). The present study shows that B-CLL cells, freshly isolated from two patients, were sIgM+, sIgG-, and sIgA- but expressed IgG and IgA transcripts. cDNA cloning and sequencing showed that the VDJ segments associated to gamma and alpha heavy chain transcripts are ident...

A new quantitative immunoperoxidase method is presented for determining absolute amounts of peroxidase and, consequently, surface antigen densities of individual cells in B lymphocytes from normal individuals, from subjects with CLL and prolymphocytic leukemia, and during ontogeny of B lympocytes in the mouse. The following results were observed: (1) The density of B antigenic sites were lower ...

Peripheral blood lymphocytes from normal donors and patients with chronic lymphocytic leukemia, B-cell type, were purified into T, helper T, and suppressor T lymphocytes by fluorescence-activated cell sorting using OKT3, OKT4, and OKT8 monoclonal antibodies. The maximum response of the purified subpopulations to stimulation by phytohemagglutinin (PHA) was determined by measuring the production ...

Background: We have previously reported the aberrant expression of Fibromodulin (FMOD) in patients with chronic lymphocytic leukemia (CLL). Although FMOD has been considered as a cytoplasmic or secretory protein, we discovered the cell surface expression of FMOD in leukemic B cells via anchoring with glycosylphosphatidylinositol (GPI). Objective: To evaluate FM...

221 samples of leukemic patients were studied, using morphologic, cytochemical staining pattern and immunophenotyping criteria. Patients included 63 children and 158 adults. Out of the adult group, 110 cases had acute leukemia including 67% AML, 330'{' ALL. In this group, 24 cases of CML" 19 cases of CLL, 4 cases of HCL and one case of plasma cell leukemia were classified. Out of 63 cases of le...

B chronic lymphocytic leukemia (B-CLL) are clonal populations of mlgM+ or mlgM+/mlgD+ CD5’ B cells that appear to be arrested in the follicular mantle-zone B-cell stage. Functional analyses have shown two groups of B-CLL that can be distinguished based on their capacity to proliferate in response to B-cell antigen receptor complex (BCRI crosslinking. To investigate the molecular basis for this ...
