The visible light spectrum is wide, and it can be hypothesized that all the wavelengths between 400-700 nm do not induce the same photobiological effects on pigmentation. We assessed the potential pro-pigmenting effects of two single wavelengths located at both extremities of the visible spectrum: the blue/violet line (λ = 415 nm) and the red line (λ = 630 nm). We made colorimetric, clinical, a...

We developed two tightly regulated, light-activated Cre recombinase enzymes through site-specific incorporation of two genetically-encoded photocaged amino acids in human cells. Excellent optical off to on switching of DNA recombination was achieved. Furthermore, we demonstrated precise spatial control of Cre recombinase through patterned illumination.

We have developed a new λ Red recombineering methodology for generating transient selection markers that can be used to transfer mutations between bacterial strains of both Escherichia coli and Salmonella enterica. The method is fast, simple and allows for the construction of strains with several mutations without any unwanted sequence changes (scar-free). The method uses λ Red recombineering t...

The λ-Red recombination system is a popular method for gene editing. However, its applications are limited due to restricted electroporation of DNA fragments. Here, we present an electroporation-free λ-Red recombination method in which target DNA fragments are excised by I-CreI endonuclease in vivo from the landing pad plasmid. Subsequently, the I-SceI endonuclease-cutting chromosome and DNA do...

We present plane-parallel and spherical LTE and NLTE atmospheric models of a variety of stellar parameters of the red giant star Arcturus (α Boo, HD124897, HR5340) and study their ability to fit the measured absolute flux distribution. Our NLTE models include tens of thousands of the strongest lines in NLTE, and we investigate separately the effects of treating the light metals and the Fe group...

A complete study of the angular distributions of the processes, Λ b → ΛV (1 −), with Λ → pπ − and V (J/Ψ) → ℓ + ℓ − or V (ρ 0 , ω) → π + π − , is performed. Emphasis is put on the initial Λ b polarization produced in the proton-proton collisions. The polarization density-matrices as well as angular distributions are derived and help to construct T-odd observables which allow us to perform tests...

The λ Red recombination system was used in this study in an attempt to inactivate the lacI gene in Escherichia coli C29 cells. The proposed model retained the first 41 amino acids of the lacI gene, and replaced the rest of the gene with a linear double-stranded DNA PCR product that confers kanamycin resistance. The PCR products encode a kanamycin cassette with lacI flanking region. The initial ...

The need for generating precisely designed mutations is common in genetics, biochemistry, and molecular biology. Here, I describe a new λ Red recombineering method (Direct and Inverted Repeat stimulated excision; DIRex) for fast and easy generation of single point mutations, small insertions or replacements as well as deletions of any size, in bacterial genes. The method does not leave any resi...

Angiotensinogen (AGT) is mainly expressed in glial cells in close proximity to renin-expressing neurons in the brain. We previously reported that glial-specific overexpression of ANG II results in mild hypertension. Here, we tested the hypothesis that glial-derived AGT plays an important role in blood pressure regulation in hypertensive mice carrying human renin (hREN) and human AGT transgenes ...
