Treatment with vanadium, a representative of a class of antidiabetic compounds, alleviates diabetic hyperglycemia and hyperlipidemia. Oral administration of vanadium compounds in animal models and humans does not cause clinical symptoms of hypoglycemia, a common problem for diabetic patients with insulin treatment. Gene expression, using Affymetrix arrays, was examined in muscle from streptozot...

K(+) currents were measured using a whole cell voltage-clamp method in enzymatically isolated rat ventricular myocytes obtained from two hyperinsulinemic, insulin-resistant models. Fructose-fed rats as well as genetically obese rats, both of which are resistant to the metabolic effects of insulin, were used. The normal augmentation of a calcium-independent sustained K(+) current was reduced or ...

Hepatic cholesterol synthesis in rabbits, as measured by the incorporation of C(14)-labelled acetate, was inhibited by addition of non-toxic amounts of vanadyl sulfate (0.05 per cent VOSO(4).2H(2)O) to the diet. This diet reduced excess aortic cholesterol in cholesterol prefed rabbits when fed during the 6 weeks immediately following a 4 week period of cholesterol feeding.

The title oxovanadium(V) complex, [V(C(18)H(13)N(3)O(2))(CH(3)O)O(CH(3)OH)], was obtained by the reaction of 1-(2-hy-droxy-naphthalen-1-yl)ethanone, nicotinohydrazide and vanadyl sulfate in methanol. The V(V) atom is six-coordinated by the N,N,O-tridentate Schiff base ligand, one methano-late O atom, one methanol O atom and one oxide O atom, forming a distorted octa-hedral geometry. The methano...

A series of chiral vanadyl carboxylates derived from N-salicylidene-L-alpha-amino acids and vanadyl sulfate has been developed. These configurationally well defined complexes were examined for the kinetic resolution of double- and mono-activated 2 degrees alcohols. The best chiral templates involve the combination of L-tert-leucine and 3,5-di-t-butyl-, 3,5-diphenyl-, or 3,4-dibromo-salicylaldeh...

Vanadyl sulfate (VOSO(4)) is an oxidative form of vanadium that in vitro and in animal models of diabetes has been shown to reduce hyperglycemia and insulin resistance. Small clinical studies of 2- to 4-week duration in type 2 diabetes (T2DM) have led to inconsistent results. To define its efficacy and mechanism of action, 11 type 2 diabetic patients were treated with VOSO(4) at a higher dose (...

Since insulin-dependent diabetes mellitus (IDDM), which causes many severe secondary complications, is characterized by hyperglyceria due to absolute deficiency of insulin, the diseases is controlled by daily injection of insulin. Therefore, the development of insulin replacements of mimetics upon oral administration is an important investigation. Recent studies indicate that vanadium, which is...

To determine whether insulin resistance and hyperinsulinemia are causally related to fructose-induced hypertension, we used vanadyl sulfate, a drug that improves insulin sensitivity in rats. Chronic oral vanadyl treatment was initiated in 6-week-old male Sprague-Dawley rats. One week after vanadyl was started, rats were fed either normal rat chow or a fructose-enriched diet. Plasma glucose and ...

The authors describe the influence of vanadyl sulphate on liver Golgi complexes in control and streptozotocin (STZ)-diabetic rats. VOSO4, one of inorganic vanadium compounds widely used in animal models and human diabetes, acts as an insulin-mimetic drug and is relatively well known as a complex activated or inhibited on many enzymes involved in carbohydrate or lipid metabolic pathways. A relat...

BACKGROUND Vanadyl sulfate (VS) and taurine are two promising agents in the treatment of diabetes related to their antihyperglycemic, antihyperlipidemic, and hyperinsulinemic effects. Data about the effects of VS on the oxidant-antioxidant system is limited and controversial. However, taurine is a well-documented antioxidant agent and our aim was to investigate the effects of VS, taurine and VS...
