We report the first complete genome sequence of a feline astrovirus (FAstV), FAstV2 strain 1637F, identified from a domestic cat. The genome is 6,779 nucleotides (nt) in length and consists of three overlapping open reading frames (ORF1a-ORF1b-ORF2). Sequence analysis suggests that FAstV2 represents a new FAstV genotype that is closely related to human astroviruses.
Using an ORF1b-based astrovirus-specfic reverse transcription (RT)-PCR assay, a novel astrovirus-like was detected from domestic geese in China. Pairwise comparisons and phylogenetic analyzes suggested that a novel group of goose astrovirus, different with previously known astroviruses in the genus Avastrovirus, was found circulating in geese. This study has expanded our understanding about the...
BACKGROUND Since the discovery of Middle East respiratory syndrome coronavirus (MERS-CoV) in 2012, diagnostic protocols were quickly published and deployed globally. OBJECTIVES We set out to assess the quality of MERS-CoV molecular diagnostics worldwide. STUDY DESIGN Both sensitivity and specificity were assessed using 12 samples containing different viral loads of MERS-CoV or common corona...
The genome of pineapple mealybug wilt-associated closterovirus-2 (PMWaV-2) was cloned from double-stranded RNA isolated from diseased pineapple and its sequence determined. The 3'-terminal 14861 nt of the single-stranded RNA genome contains ten open reading frames (ORFs) which, from 5' to 3', potentially encode a >204 kDa polyprotein containing papain-like protease, methyltransferase and helica...
The self-replicating polerovirus-associated RNAs have recently been classified as tombusvirus-like associated (tlaRNAs). In a metagenomic comparison of the virome wild (Daucus carota subsp. carota) and cultivated (D. sativus) carrots, four tlaRNAs were identified: carrot red leaf virus-associated RNA (CtRLVaRNA), beet western yellows (BWYVaRNA), two other detected for first time in arracacha la...
conclusions this study showed that the developed rrt-pcr assays are rapid, reliable, reproducible, specific, sensitive, and simple tools for detection of mers-cov. finally, a kit consisting of two assay signatures and controls was assembled, which can be distributed to public health laboratories in iran to support international mers-cov surveillance and public health response. results the upe a...
Detection of a novel human coronavirus by real-time reverse-transcription polymerase chain reaction.
We present two real-time reverse-transcription polymerase chain reaction assays for a novel human coronavirus (CoV), targeting regions upstream of the E gene (upE) or within open reading frame (ORF)1b, respectively. Sensitivity for upE is 3.4 copies per reaction (95% confidence interval (CI): 2.5–6.9 copies) or 291 copies/mL of sample. No cross-reactivity was observed with coronaviruses OC43, N...
Nidoviruses with large genomes (26.3-31.7 kb; 'large nidoviruses'), including Coronaviridae and Roniviridae, are the most complex positive-sense single-stranded RNA (ssRNA+) viruses. Based on genome size, they are far separated from all other ssRNA+ viruses (below 19.6 kb), including the distantly related Arteriviridae (12.7-15.7 kb; 'small nidoviruses'). Exceptionally for ssRNA+ viruses, large...
