Corynebacterium efficiens is the closest relative of Corynebacterium glutamicum, a species widely used for the industrial production of amino acids. C. efficiens but not C. glutamicum can grow above 40 degrees C. We sequenced the complete C. efficiens genome to investigate the basis of its thermostability by comparing its genome with that of C. glutamicum. The difference in GC content between t...
Bacteria regulate chromosome replication and segregation tightly with cell division to ensure faithful segregation of DNA to daughter generations. The underlying mechanisms have been addressed in several model species. It became apparent that bacteria have evolved quite different strategies to regulate DNA segregation and chromosomal organization. We have investigated here how the actinobacteri...
The transcriptional regulator Cg1486 of Corynebacterium glutamicum ATCC 13032 is a member of the IclR protein family and belongs to the conserved set of regulatory proteins in corynebacteria. A defined deletion in the cg1486 gene, now designated ltbR (leucine and tryptophan biosynthesis regulator), led to the mutant strain C. glutamicum IB1486. According to whole-genome expression analysis by D...
background: l-glutamic acid is one of the major amino acids that is present in a wide variety of foods. it is mainly used as a food additive and flavor enhancer in the form of sodium salt. corynebacterium glutamicum (c. glutamicum) is one of the major organisms widely used for glutamic acid production. methods: the study was dealing with immobilization of c. glutamicum and mixed culture of c. g...
Mycolic acids, the major lipid constituents of Corynebacterineae, play an essential role in maintaining the integrity of the bacterial cell envelope. We have previously characterized a corynebacterial mycoloyltransferase (PS1) homologous in its N-terminal part to the three known mycobacterial mycoloyltransferases, the so-called fibronectin-binding proteins A, B and C. The genomes of Corynebacte...
A protoplast transformation system has been developed for Corynebacterium glutamicum by using a C. glutamicum-Bacillus subtilis chimeric vector. The chimera was constructed by joining a 3.0-kilobase cryptic C. glutamicum plasmid and the B. subtilis plasmid pBD10. The neomycin resistance gene on the chimera, pHY416, was expressed in C. glutamicum, although the chloramphenicol resistance gene was...
BACKGROUND Corynebacterium glutamicum was used as a metabolic engineering chassis for production of crude violacein (mixture of violacein and deoxyviolacein) due to Corynebacterium's GRAS status and advantages in tryptophan fermentation. The violacein is a commercially potential compound with various applications derived from L-tryptophan. RESULTS Corynebacterium glutamicum ATCC 21850 that co...
The promoters and plasmids of Corynebacterium glutamicum can be viewed as subjects for genetic examination, tools for studies of gene expression, and a means for the improvement of industrial strains. Various classes of C. glutamicum promoters, their specific functions in gene expression control, and methods for their analysis are described in this review. C. glutamicum promoters recognized by ...
Direct polyhydroxybutyrate (PHB) production from starch was for the first time achieved using engineered Corynebacterium glutamicum expressing PHB biosynthetic genes and displaying α-amylase on its cell surface. The engineered strain accumulated 6.4 wt% PHB from starch which was higher than that obtained from glucose (4.9 wt%).
Background: Corynebacterium glutamicum (C. glutamicum) is a potential host for the secretory production of the heterologous proteins. However, to this date few secretion-type gene expression systems in C. glutamicum have been developed, which limit applications of C. glutamicum in a secretory production of the heterologous proteins. Objectives: In this stu...
