Vaccinia virus replicates in the cytoplasm of the host cell and encodes its own RNA polymerase and transcription factors. The proteins that target the poxvirus RNA polymerase to intermediate- and late-class promoters have not been identified. In this study, representatives of the intermediate and late promoters were characterized at the nucleotide level to identify essential motifs. Both interm...
The TATA box binding protein (TBP) plays a central and essential role in transcription initiation. At TATA box-containing genes transcribed by RNA polymerase II, TBP binds to the promoter and initiates the assembly of a multiprotein preinitiation complex. Several studies have suggested that binding of TBP to the TATA box is an important regulatory step in transcription initiation in vitro. To d...
This study designed, techno-economically analyzed and compared the heat-assisted extraction (HAE)- ultrasound-assisted (UAE)-technology-based processes (HAE-tBP UAE-tBP) for production of crude anthocyanins powder (CAnysP) from Hibiscus sabdariffa calyx (HSC) with assistance SuperPro Designer software. Both base cases industrial setups 600 L extractor capacities HAE- UAE-tBPs were compared. Whe...
INTRODUCTION Tuberculosis peritonitis (TBP) is uncommon and symptoms can be non-specific. Local data are lacking and our aim is to review our local experience with TBP. MATERIALS AND METHODS Between the period of 1996 to 2004, there were 10 [male: 6; mean age, 48 +/- 18.5 years (range, 26 to 73)] cases of histologically documented TBP. Comparisons were made with pulmonary tuberculosis (PTB) p...
Simian virus 40 (SV40) large tumor (T) antigen is the major regulatory protein that directs the course of viral infection, primarily by interacting with host cell proteins and modulating their functions. Initiation of viral DNA replication requires specific interactions of T antigen bound to the viral origin of DNA replication with cellular replication proteins. Transcription factors are though...
Regulation of transcription involves the activities of activators and repressors. Recent experiments have provided evidence that the function of both types of regulators can involve interactions with one or more component of the basal transcription machinery. A principal target appears to be TFIID, which consists of the TATA binding protein (TBP) and associated factors (TAFs). Here we describe ...
Previous work has shown that binding of the TATA box binding protein (TBP) to the TATA box is a rate-limiting step during pre-initiation complex (PIC) formation. Although the transcription of eukaryotic genes normally proceeds in one direction, studies in solution have shown that TBP lacks the information necessary to orient itself on the TATA box. Instead, yeast TBP binds TATA-containing promo...
Transcription initiation in Archaea requires the assembly of a preinitiation complex containing the TATA- box binding protein (TBP), transcription factor B (TFB), and RNA polymerase (RNAP). The results reported establish the fate of Methanothermobacter thermautotrophicus TBP and TFB following transcription initiation by M. thermautotrophicus RNAP in vitro. TFB is released after initiation, duri...
Transcription factors TBP and TF(II)B assemble with RNA polymerase at the promoter DNA forming the initiation complex. Despite a high degree of conservation, the molecular binding mechanisms of archaeal and eukaryotic TBP and TF(II)B differ significantly. Based on recent biophysical data, we speculate how the mechanisms co-evolved with transcription regulation and TBP multiplicity.
BACKGROUND Tuberculous pericarditis (TBP) is associated with high morbidity and mortality, and is an important treatable cause of heart failure in developing countries. Tuberculous aetiology of pericarditis is difficult to diagnose promptly. The utility of the new quantitative PCR test (Xpert MTB/RIF) for the diagnosis of TBP is unknown. This study sought to evaluate the diagnostic accuracy of ...
