Astrocytes secrete cholesterol in lipoprotein particles. Here we show that primary murine embryonic astrocytes secrete endogenously synthesized cholesterol but also the cholesterol precursors desmosterol and lathosterol. In astrocyte membranes, desmosterol and cholesterol were the predominant sterols. Astrocytes derived from Niemann-Pick type C lipidosis (NPC1-/-) mice displayed late endosomal ...

BACKGROUND Global sterol analysis is challenging owing to the extreme diversity of sterol natural products, the tendency of cholesterol to dominate in abundance over all other sterols, and the structural lack of a strong chromophore or readily ionized functional group. We developed a method to overcome these challenges by using different isotope-labeled versions of the Girard P reagent (GP) as ...

Apomine, a novel 1,1-bisphosphonate ester, has been shown to lower plasma cholesterol concentration in several species. Here we show that Apomine reduced the levels of 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMGR), the rate-limiting enzyme in the mevalonate pathway, both in rat liver and in cultured cells. Apomine resembles sterols such as 25-hydroxycholesterol in its ability to potent...

A Tortricidae (Lobesia botrana) has a mutualistic relationship with the fungus (Botrytis cinerea). In this study, we investigated the growth, survival, fecundity and amount of sterols and steroids in larvae of this vineyard pest reared on artificial diets containing mycelium (3%) or purified sterols (0.01%) of the phytopathogenic fungus. Two principal questions related to the physiological and ...

The ATP binding cassette transporters ABCG5 (G5) and ABCG8 (G8) limit the accumulation of neutral sterols by restricting sterol uptake from the intestine and promoting sterol excretion into bile. Humans and mice lacking G5 and G8 (G5G8-/-) accumulate plant sterols in the blood and tissues. However, despite impaired biliary cholesterol secretion, plasma and liver cholesterol levels are lower in ...

Cholesterol transport between cellular organelles comprised vesicular trafficking and nonvesicular exchange; these processes are often studied by quantitative fluorescence microscopy. A major challenge for using this approach is producing analogs of cholesterol with suitable brightness and structural and chemical properties comparable with those of cholesterol. This review surveys currently use...

The effects of amidated carboxymethylcellulose, amidated pectin, and psyllium on serum and hepatic cholesterol, hepatic fat, and fecal output of sterols were examined in female rats. Rats were fed a diet supplemented with cholesterol (0 or 10 g/kg) and palm fat. Amidated cellulose at 30 g/kg significantly decreased the serum and hepatic concentration of cholesterol by 28.1% and 64.6%, respectiv...

Following a recent experimental investigation of the effect of the length of the alkyl side chain in a series of cholesterol analogues (Angew. Chem., Int. Ed., 2013, 52, 12848-12851), we report here an atomistic molecular dynamics characterization of the behaviour of methyl-branched side chain sterols (iso series) in POPC bilayers. The studied sterols included androstenol (i-C0-sterol) and chol...

Caenorhabditis elegans requires sterol, usually supplied as cholesterol, but this is enzymatically modified, and different sterols can substitute. Sterol deprivation decreased brood size and adult growth in the first generation, and completely, reversibly, arrested growth as larvae in the second. After one generation of sterol deprivation, 10 ng/ml cholesterol allowed delayed laying of a few eg...

We performed a meta-analysis of five randomized, placebo-controlled trials to characterize the impact of plant sterols/stanols on plasma lipids in patients with type 2 diabetes. Upon meta-analysis, plant sterols/stanols significantly reduced total and LDL cholesterol, with a trend towards improvement in HDL. No beneficial effect on triglycerides was apparent.
