The slow folding of circularly permuted variants of ribonuclease T1 has been examined using steady-state and frequency-domain fluorescence spectroscopy. The sequence transpositions have previously been designed by eliminating a restrictive Cys2-Cys10 disulfide bond, adjoining the original termini with a three-peptide Gly-Gly-Gly linker, and conferring new termini to four different solvent-expos...

Sequential disassembly (MSn) has been applied to fully characterise and document native samples containing glycan epitopes with their synthetic analogues. Both sample types were prepared by methylation, solvent phase extracted, directly infused and spatially resolved. Product ions of all samples were compiled and contrasted using management tools prepared for the fragment ion library. Each of t...

The RNA cleavage reaction catalyzed by the hairpin ribozyme shows biphasic kinetics, and chase experiments show that the slow phase of the reaction results from reversible substrate binding to an inactive conformational isomer. To investigate the structural basis for the heterogeneous kinetics, we have developed an enzymatic RNA modification method that selectively traps substrate bound to the ...

The title compound, C(19)H(19)NO(2), obtained as an almost equimolar mixture (as shown by (1)H NMR) with the E isomer through a Wittig reaction between 4-amino-9H-fluoren-9-one and the stabilized ylide Ph(3)P=CHCO(2)C(CH(3))(3), was obtained pure in the Z configuration following crystallization from toluene. The mol-ecule shows a planar arrangement of the ring system and the new double bond, wh...

Five children with atrial isomerism developed intestinal obstruction caused by malrotation of the gut. Other than asplenia, the extracardiac anomalies in these syndromes are rarely regarded as important as the outcome after intestinal surgery is poor. As cardiac treatment improves, early investigation and intervention for intestinal symptoms becomes more important.

Electronic structure calculations employing density functional theory (DFT) and time-dependent density functional theory (TD-DFT) have been carried out on the model complexes {[(HCO2)3M2]2(mu-O2CCO2)}0/+(M=Mo or W) in D2h symmetry, where the oxalate bridge forms either five- or six-membered rings with the M(2) centres; the complexes are hereafter referred to as mu(5,5)0/+ and mu(6,6)0/+, respec...

Photoresponsive systems for site-selective RNA scission have been prepared by combining Lu(III) ions with acridine/azobenzene dual-modified DNA. The modified DNA forms a heteroduplex with substrate RNA, and the target phosphodiester linkages in front of the acridine residue is selectively activated so that Lu(III) ion rapidly cleaves the linkage. Azobenzene residue introduced adjacent to the ac...

Structural requirements of N-blocked L-proline derivatives as specific inhibitors for prolyl endopeptidase were investigated using a series of substrate analogs. Replacement of L-proline by its D-isomer remarkably reduced the inhibition. Introduction of a sulfur atom in proline and/or in the penultimate pyrrolidine rings significantly increased the inhibition, but the introduction of oxygen rat...

A yellow chromophore has been isolated from Avena coleoptiles grown in the dark. It had previously been shown by Zenk [Zenk, M. H. (1967) Z. Pflanzenphysiol. 56, 122 - 140] to be a flavin of still unidentified structure, and had been suggested to be the 'blue light' photoreceptor in this organism. The structure of this flavin (F1-X) has been identified as 5'-malonylriboflavin by a combination o...

We report the case of an asymptomatic six-year-old child with left atrial isomerism and sinus venosus atrial septal defect. The physical examination revealed several periods of bradycardia. During a 24-hour electrocardiographic monitoring the patient presented a significant sinus node dysfunction with sinus pauses of up to 2.4 seconds. A permanent pacemaker was implanted, with a satisfactory ou...
