In June of 1987, an outbreak of penicillinase-producing Neisseria gonorrhoeae (PPNG) occurred in Rhode Island. PPNG persists as an endemic pathogen despite a concerted statewide effort to eradicate the organism. Detailed analysis of PPNG isolates demonstrated that multiple strains are circulating concurrently, complicating control measures.

A multitarget PCR was developed for the direct detection of penicillinase-producing Neisseria gonorrhoeae (PPNG). The assay was validated by testing 342 PPNG isolates and 415 clinical samples. The method is suitable for routine detection of PPNG strains. Its multitarget approach reduces the potential for false-negative results caused by sequence variations.

The in vitro susceptibility of penicillinase-producing and intrinsically resistant Neisseria gonorrhoeae strains to 13 antimicrobial agents was tested. Regardless of the type of resistance, these organisms remained quite susceptible to newer cephalosporin agents, including moxalactam, cefoperazone, cefotaxime, and ceftazidime.
