We have discovered a novel polyomavirus present in multiple human stool samples. The virus was initially identified by shotgun pyrosequencing of DNA purified from virus-like particles isolated from a stool sample collected from a healthy child from Malawi. We subsequently sequenced the virus' 4,927-bp genome, which has been provisionally named MW polyomavirus (MWPyV). The virus has genomic feat...

We measured the accumulation of virus-specific early RNA in BALB/3T3 cells infected by the ts3 mutant of polyoma virus by annealing cytoplasmic RNA from infected cells to purified, radiolabeled, "early" strand of polyoma DNA. Cells infected by the ts3 mutant fail to accumulate virus-specific early RNA at the nonpermissive temperature.

Merkel cell carcinoma (MCC) is an aggressive, polyomavirus-associated skin cancer. Robust cellular immune responses are associated with excellent outcomes in patients with MCC, but these responses are typically absent. We determined the prevalence and reversibility of major histocompatibility complex class I (MHC-I) downregulation in MCC, a potentially reversible immune-evasion mechanism. Cell-...

Approximately 15-20% of human cancer is related to infection, which renders them potentially preventable by antimicrobial or antiviral therapy. Human polyomaviruses (PyVs) are relevant in this regard, as illustrated by the involvement of Merkel cell polyomavirus (MCPyV) in the development of Merkel cell carcinoma. The polyomavirus Small and Large tumor antigen (ST and LT) have been extensively ...

The large T antigen (TAg) of the polyomavirus Simian virus 40 (SV40) is known to play an important role in several events during the viral life cycle, including DNA replication, transcription, and virion assembly. Recent analysis revealed that primary mouse embryonic fibroblasts (MEFs) expressing exogenous TAg showed increased transcription of interferon stimulated genes (ISGs). Interestingly, ...

Polyomavirus-associated nephropathy in kidney transplantation is viewed as a complication of prolonged, intense immunosuppression, which disrupts the balance between antiviral immune control and polyomavirus replication. The prevalence rate ranges from 1-10% in kidney transplant programs around the world, with graft failure and return to dialysis in > 50%. Most cases are caused by the human pol...
