In order to investigate SAR regarding proximal phenyl ring in novel C-aryl glucoside SGLT2 inhibitors containing a thiazole motif, a series of chemical modifications on proximal phenyl ring was conducted. This work describes the design and application of a single-pot, reductive arene C-H silanolization of aromatic esters for synthesis of ortho-formyl arylsilanols. This strategy involves a sequence of two transition metal (Ir and Rh)-catalyzed reactions for reductive arene ortho-silylation directed by hydridosilyl acetals and hydrolysis.

The highly regioselective ortho-arylation of acetanilides with aromatic boronic acids in the presence of a Ru(II) complex (3 mol%), AgSbF6 (12 mol%), Cu(OTf)2 (20 mol%) and Ag2O (1.0 eq.) is described. Later, ortho-arylated acetanilides were converted into phenanthridine and carbazole derivatives by using Ph3PO and Tf2O or palladium or Cu(OTf)2 catalysts.
