A point mutation (E115K) resulting in slower growth of Escherichia coli DH5alpha and XL1-Blue in minimal media was identified in the purB gene, coding for adenylosuccinate lyase (ASL), through complementation with an E. coli K-12 genomic library and serial subcultures. Chromosomal modification reversing the mutation to the wild type restored growth phenotypes in minimal media.

No statistical differences were found in the recovery of group A streptococci from throat culture specimens after overnight incubation of blood agar plates in 5% CO2 compared with anaerobiosis. Anaerobic incubation required many more subcultures and resulted in considerably greater technical time and expense.

A study of 16,108 blood cultures investigated the value of performing early smears in addition to the inspection of early subcultures after 12 h of incubation. The early smear and subculture detected 44.6% of 1,236 positive cultures; of those detected by the early subculture, 57.3% grew after 12 h of incubation.

The ability of 50 sequential subcultures in subinhibitory concentrations of telithromycin (HMR 3647), azithromycin, clarithromycin, erythromycin A, roxithromycin, clindamycin, and pristinamycin to select for resistance was studied in five macrolide-susceptible and six macrolide-resistant pneumococci containing mefE or ermB. Telithromycin selected for resistance less often than the other drugs.
