長年，アレルゲンの解析手法としてSDS-PAGEとWestern blottingが用いられてきた．この場合，タンパク質をできるだけ可溶化するために，SDSなどの変性剤を高濃度で添加することが多い．筆者らはこの手法を用いて，甲殻類アレルゲンの種特異的反応性の違いを検出した．ところが，別の患者血清では，この方法ではバンドが全く検出されなかった．そこでこの患者血清をdot blottingで調べたところ，生では強く反応したが，80°Cで15分加熱すると反応が失われた．そこで，生の試料をdot blotしたPVDF膜を0.1％SDSを含む電気泳動バッファー中で1時間インキュベートしたところ反応性は弱まったが，完全には反応が消失していなかった．そこで，0.1％SDSを含むSDS抽出液を用いてSDS-PAGEを行ったところ，トロポミオシンのバンドが弱く反応した．甲殻類アレルゲンのSDS変性を低...
