Lysosomal neuraminidase (sialidase) occurs in a high molecular weight complex with the glycosidase beta-galactosidase and the serine carboxypeptidase protective protein/cathepsin A (PPCA). Association of the enzyme with PPCA is crucial for its correct targeting and lysosomal activation. In man two genetically distinct storage disorders are associated with either a primary or a secondary deficie...

A molecular model of the swine influenza A/H1N1 type-I neuraminidase was built using the pathogenic avian H5N1 type-I neuraminidase as a basis, due to the higher sequence identity between A/H1N1 and H5N1 (91.47%) compared to Spanish H1N1 (88.37%) neuraminidase. All-atom molecular dynamics (MD) simulations of all three neuraminidases were performed, either as apo-structures or with commercial an...

Antigenic relationships between an isolate (B/Aichi/I/73) from the epidemic of influenza B in 1973 and earlier strains of the same type were studied by hemagglutination-inhibition and neuraminidase-inhibition tests. The results showed that B/Aichi/I/73 underwent considerable antigenic variation in hemagglutinin but not in neuraminidase.

In February 2016, three influenza B/Victoria/2/87 lineage viruses exhibiting 4- to 158-fold reduced inhibition by neuraminidase inhibitors were detected in Laos. These viruses had an H134N substitution in the neuraminidase and replicated efficiently in vitro and in ferrets. Current antiviral drugs may be ineffective in controlling infections caused by viruses harboring this mutation.

A fluorometric substrate, 4-methylumbelliferyl-alpha-ketoside of N-acetylneuramide, was used directly on clinical specimens and infected tissue culture 24 h after inoculation for the detection of influenza viral neuraminidase. Viral neuraminidase was detected in infected tissue culture but not in clinical specimens. The sensitivity of the assay on tissue culture was 92%, and the specificity was...

To identify oseltamivir resistance, we analyzed neuraminidase H275Y mutations in samples from 10 patients infected with pandemic (H1N1) 2009 virus in South Korea who had influenza that was refractory to antiviral treatment with this drug. A neuraminidase I117M mutation that might influence oseltamivir susceptibility was detected in sequential specimens from 1 patient.
