Photosensitizing flavoproteins have great potential as tags for correlative light and electron microscopy (CLEM). We examine the photostability of miniSOG mutants and their ability to photo-oxidize diaminobenzidine, both key aspects for CLEM. Our experiments reveal a complex relation between these parameters and the production of different reactive oxygen species.
M-DPT, a BODIPY-based water soluble near-infrared fluorescent probe with thiophene at the 1,7-position, is synthesized. M-DPT is found to possess high specificity to mitochondria, superior photostability, and appreciable tolerance to microenvironmental changes. In triplet-triplet annihilation-based photon upconversion (TTA-UC), the utilization of near-infrared (NIR) light with a wavelength longer than 850 nm remains an outstanding issue. We realized this by employing metallonaphthalocyanines as triplet sensitizers; upon excitation of NIR light (856 nm), upconverted emission was observed in the visible range with remarkable photostability.
