The reaction of cis-diamminedichloroplatinum(II) and several synthetic or natural double-stranded polydeoxyribonucleotides has been carried out in the presence of such intercalating agents as ethidium bromide, proflavine, and acridine. After incubation of the reaction mixtures at 37 degrees C for 24 hr, some ethidium or proflavine, but no acridine, molecules are tightly bound to nucleic acids. ...

Reactions of the anionic gallium(i) heterocycle, [:Ga{[N(Ar)C(H)](2)}](-) (Ar = C(6)H(3)Pr(i)(2)-2,6), with a variety of mono- and bidentate phosphine, tmeda and 1,5-cyclooctadiene (COD) complexes of group 10 metal dichlorides are reported. In most cases, salt elimination occurs, affording either mono(gallyl) complexes, trans-[MCl{Ga{[N(Ar)C(H)](2)}}(PEt(3))(2)] (M = Ni or Pd) and cis-[PtCl{Ga{...

Previous studies have shown that exposure of He La cells stably transfected with an HIV-long terminal repeat-chloramphenicol acetyltransferase (HIV-LTR-CAT) construct to many DNA-damaging agents (such as UV light) induces expression from the HIV LTR. By culturing the cells with salicylic acid we demonstrated dose-dependent repression of this IVor c/'s-platinum (m-Pt)-induced HIV expression. Whi...

A sequence of stages in the syntheses of isomeric bisamino acid complexes of Pt(II) with beta-aminopropionic acid (beta-alanine = beta-AlaH) has been studied by the (195)Pt NMR spectroscopy. The techniques have been developed of the synthesis of the cis- and trans-bischelates of Pt(II) and Pd(II) with beta-alanine as well as of the halide complexes of trans-[M(beta-AlaH)(2)Cl(2)] (M = Pt, Pd) a...

A new platinum derivative of the form H[Pt(IV)(Hdigly)Cl(2)(OH)(2)] (Hdigly==glycylglycine) damaged the Candida albicans cell membrane and inhibited the growth of the cells. The cytotoxic activity of H[Pt(IV)(Hdigly)Cl(2)(OH)(2)] on mammalian cells was 10-fold lower than that of cis-diammine-dichloroplatinum (cisplatin). Substitution of platinum for peptides is effective for enhancement of anti...

The HSQC NMR spectrum of 15N-cisplatin in cell growth media shows resonances corresponding to the monocarbonato complex, cis-[Pt(NH3)2(CO3)Cl](-), 4, and the dicarbonato complex, cis-[Pt(NH3)2(CO3)2](-2), 5, in addition to cisplatin itself, cis-[Pt(NH3)2Cl2], 1. The presence of Jurkat cells reduces the amount of detectable carbonato species by (2.8+/-0.7) fmol per cell and has little effect on ...

In the crystal structure of the title complex, [Pt(C(5)H(4)NOS)(2)], the Pt atom is coordinated by two O atoms and two S atoms in a cis configuration, forming a distorted square-planar coordination geometry. The mol-ecule exhibits pseudo-C(2v) symmetry and is essentially planar, with a maximum deviation from planarity of 0.0124 (2) Å. The dihedral angle between the two pyridine rings is 5.85 (2)°.

Cisplatin is one of the most successful compounds in the fight against cancer. Recently, interest was directed towards the development of cisplatin analogues which possess Nheterocyclic carrier ligands, coordinated to the cytotoxic platinum(II) moiety, instead of one or both of the am(m)ines. Hydantoins form a large group of derivatives widely applied in medicine and pharmacy, especially as ant...

Cell killing and specificity of mutation induction by treatment of Escherichia coli K-12 cells with cis-Pt(NH3)2Cl2 at various temperatures have been studied. Survival experiments show that the cell killing by cis-Pt(NH3)2Cl2 is enhanced with increasing temperature. This effect is explained by an increase in the amount of platinum bound to the DNA. The binding is the same in repair-proficient a...

The title compound, [PtCl(2)(C(25)H(21)N(2)P)], is a Pt(II) complex with an NPCl(2) coordination sphere in which the metal is coordinated to the imino N and phosphane P atoms of the ligand and to two chloride ions. The Pt(II) atom is in a distorted square-planar environment and is bound to the ligand via the P and amine N atoms in a cis fashion, with the chlorine atoms located at the two remain...
