This paper describes the overnight Giemsa staining method for detecting mycoplasma infection of cell cultures. This simple method requires no special equipment and has practical application in most cell culture laboratories. This technique has been used conveniently to monitor stock cell cultures for mycoplasma infection over a 12-month period avoiding any contamination.

A direct Giemsa technique is described for the selective staining of Brd-U-substituted chromatin. It can advantageously be used for sister-chromatid exchange analysis and for the detailed cytological study of the mouse kinetochores. Presumptive inversions within the centromeric region of chromosomes 4 and 18 in the C57Black/6J mouse are described.

The bibenzimidazol derivative 33258 Hoechst can be used to distinguish microfluorometrically between mouse and human nuclei in heterokaryons. This affords a quick and accurate alternative to autoradiography in the analysis of such heterokaryons. The 33258 Hoechst fluorescence patterns can be converted after irradiation to a Giemsa rendition of the differential staining.

An exponential population of Saccharomyces cerevisiae cells was fractionated by centrifugal elutriation, using water as the elutriating liquid. Evidence that the population had been fractionated according to age in the cell cycle was obtained by examining the fractions for their size distribution, their microscopic appearance after Giemsa staining, and their ability to initiate synchronous growth.
