A closed-tube, real-time PCR assay was developed for sensitive and specific detection and differentiation of the two closely related intestinal protozoan parasites Entamoeba histolytica and Entamoeba dispar directly from human feces. The assay is performed with the LightCycler system using fluorescence-labeled detection probes and primers amplifying a 310-bp fragment from the high-copy-number, ...

INTRODUCTION Entamoeba histolytica infections were investigated in residents of the Ariquemes and Monte Negro municipalities in Rondônia State, Brazil. METHODS Stool samples of 216 individuals were processed by the spontaneous sedimentation method and analyzed by microscopy for detection of the E. histolytica/E. dispar complex, followed by the immunoassay method using an enzyme-linked immunos...

OBJECTIVE Humans may be infected with three morphologically identical Entamoeba species; pathogenic E. histolytica, commensal E. moshkowskii and E. dispar. The aim of the present study was to determine the true prevalence of the E. histolytica using native lugol, trichrome staining and a monoclonal antigen detection kit (ELISA kit E. histolytica-II; Techlab, Inc., Blacksburg, VA) among primary ...

Background: Entamoeba (E.) histolytica, is a protozoan parasite that causes intestinal and extraintestinal amebiasis, leading to significant morbidity mortality in developing nations. E. dispar, moshkovskii, members of E.histolytica complex species, are all morphologically similar. Aim the study: The current work aimed utilize molecular techniques for identifying exact prevalence histolytica sp...

Specific identification of Entamoeba spp. in clinical specimens is an important confirmatory diagnostic step in the management of patients who may be infected with Entamoeba histolytica, the species that causes clinical amebiasis. Distinct real-time PCR protocols have recently been published for identification of E. histolytica and differentiation from the morphologically identical nonpathogeni...

Many microscopic-based epidemiological surveys on the prevalence of human intestinal pathogenic and non-pathogenic protozoa including intestinal amoeba performed in Iran show a high prevalence of human intestinal amoeba in different parts of Iran. Such epidemiological studies on amoebiasis are confusing, mainly due to recently appreciated distinction between the Entamoeba histolytica, E. dispar...

The objective this study was to determine the occurrence of the species Entamoeba histolytica/Entamoeba díspar in clinical samples of ambulatory patients in Pernambuco. A specific assay for Entamoeba histolytica was used in this study, which identified no positive fecal samples among the 213 patients. These results confirm that E. dispar is the dominant species in Pernambuco State.

For many years, microscopic examination of stool samples has been considered to be the “gold standard” for diagnosis of intestinal parasites although the Polymerase Chain Reaction (PCR) analysis is increasingly utilized due to its high accuracy. Recently, PCR has been approved by the World Health Organization as the current method of choice for the diagnosis of Entamoeba histolytica infection. ...

BACKGROUND Entamoeba moshkovskii and E. dispar are impossible to differentiate microscopically from the pathogenic species E. histolytica. Multiplex polymerase chain reaction (Multiplex PCR) is a widespread molecular biology technique for amplification of multiple targets in a single PCR experiment. METHODS For detection and differentiation of the three-microscopy indistinguishable Entamoeba ...

Genes coding for human antibody Fab fragments specific for Entamoeba histolytica were cloned and expressed in Escherichia coli. Lymphocytes were separated from the peripheral blood of a patient with an amebic liver abscess. Poly(A)+ RNA was isolated from the lymphocytes, and then genes coding for the light chain and Fd region of the heavy chain were amplified by a reverse transcriptase PCR. The...
