BACKGROUND Only 2 allergenic proteins have been described in lettuce allergy: a 16-kDa protein (putative profilin) and a lipid transfer protein (LTP) named Lac s 1. OBJECTIVE Our aim was to identify the allergens involved in the anaphylactic reactions of 2 patients who had eaten lettuce. METHODS The study was performed by Ig (immunoglobulin)-E immunodetection and immunodetection-inhibition ...

The orientational texture of gel-phase lipid bilayers is a phenomenon that can structure membrane domains. Using two-photon polarized fluorescence microscopy and image analysis, we map the lateral variation of the lipid orientation (the texture) in single domains. With this method, we uncover a lipid-induced transition between vortex and uniform textures in binary phospholipid bilayers. By tuni...

In this paper we show the feasibility on using silicon batch-fabricated non planar microsized aperture for the monitoring of membrane proteins inserted in a lipid bilayer reconstituted on this aperture. A low cost chemical etching protocol was used for micromachining the pore. This work is a first step towards integrated silicon biochips designed for membrane protein recording.

Leptospira interrogans is a spirochete that is responsible for leptospirosis, a zoonotic disease. This bacterium possesses an unusual LPS that has been shown to use TLR2 instead of TLR4 for signaling in human cells. The structure of its lipid A was recently deciphered. Although its overall hexa-acylated disaccharide backbone is a classical feature of all lipid A forms, the lipid A of L. interro...

The interactions of the basic, cell-penetrating region (Y47GRKKRRQRRR57) of the HIV-1 Tat protein with dioleoylphosphatidylcholine (DOPC) bilayers were previously assessed by comparing experimental X-ray diffuse scattering with atomistic molecular dynamics simulations. Here, we extend this investigation by evaluating the influence of phosphatidylethanolamine (PE) lipids. Using experimental bila...

We show that an electric field can drive single-stranded RNA and DNA molecules through a 2.6-nm diameter ion channel in a lipid bilayer membrane. Because the channel diameter can accommodate only a single strand of RNA or DNA, each polymer traverses the membrane as an extended chain that partially blocks the channel. The passage of each molecule is detected as a transient decrease of ionic curr...

The plasma membrane of human red blood cells consists of a lipid bilayer attached to a regular network of underlying cytoskeletal polymers. We model this system at a dynamic coarse-grained level, treating the bilayer as an elastic sheet and the cytoskeletal network as a series of phantom entropic springs. In contrast to prior simulation efforts, we explicitly account for dynamics of the cytoske...

Haptotaxis is a mechanism proposed at the end of the 1960s to explain cell motility. It describes cell movement induced by an adhesion gradient. In this work, we present evidence for self-induced haptotaxis using negatively charged giant vesicles interacting with positively charged supported lipid bilayers, which has not been previously described. Depending on the charge of the vesicle, we obse...

In order to investigate structural and dynamical properties of local anesthetic articaine in a model lipid bilayer, a series of molecular dynamics simulations have been performed. Simulations were carried out for neutral and charged (protonated) forms of articaine inserted in fully hydrated dimyristoylphosphatidylcholine (DMPC) lipid bilayer. For comparison purpose, a fully hydrated DMPC bilaye...

Lipid rafts are liquid-ordered membrane microdomains that form by preferential association of 3-β-hydroxysterols, sphingolipids and raft-associated proteins often having acyl modifications. We isolated lipid rafts of the protozoan parasite Trypanosoma brucei and determined the protein composition of lipid rafts in the cell. This analysis revealed a striking enrichment of flagellar proteins and ...
