We report the development of a flavivirus immunoglobulin M (IgM) capture enzyme-linked immunosorbent assay (MAC-ELISA) which improves the determination of an infecting flavivirus serotype over that by current serological methods. A panel of 165 IgM-positive sera from flavivirus patients with specific diagnostic results was tested by the flavivirus MAC-ELISA using a panel of 10 antigens. For 134...

The hemagglutination-inhibition (HI) test for antirhinovirus antibody was carried out on paired sera from volunteers inoculated with rhinovirus type 3 or type 4 (RV4). The HI test gave results which paralleled the neutralization test and was at least as sensitive as a microneutralization method for detection of serotype-specific antibody. Although high levels of HI antibody in the serum were as...

Eight strains of a new intermediate adenovirus were isolated in the course of an investigation of conjunctivitis in Hiroshima City, Japan. The strain was first isolated in July 1986. All eight strains were isolated from conjunctival swab samples from patients with conjunctivitis, mainly epidemic keratoconjunctivitis. The virus was typed as adenovirus type 22 in neutralization tests and was rela...

Nonspecific inhibitors of rubella hemagglutination can be removed by treatment of sera with heparin-manganous chloride for use in the hemagglutination-inhibition test. After removal of nonspecific inhibitors by this procedure, an excess of manganous chloride may remain. This may cause the cells to agglutinate, thus obscuring the reading at low serum dilutions. This disadvantage can be overcome ...

Indirect hemagglutination inhibition tests were employed to detect slime in concentrations as low as 1 mug/ml. Increasing concentrations of slime resulted in proportionately greater inhibition of hemagglutination. Peritoneal aspirates and plasma of mice injected with slime were shown to exhibit the inhibitory activity of slime. The rapid dissemination of slime into the peripheral circulation wa...

Antigenic relationships between an isolate (B/Aichi/I/73) from the epidemic of influenza B in 1973 and earlier strains of the same type were studied by hemagglutination-inhibition and neuraminidase-inhibition tests. The results showed that B/Aichi/I/73 underwent considerable antigenic variation in hemagglutinin but not in neuraminidase.

Recombinant soluble trimeric influenza A virus (IAV) hemagglutinin (sHA(3)) has proven an effective vaccine antigen against IAV. Here, we investigate to what extent the glycosylation status of the sHA(3) glycoprotein affects its immunogenicity. Different glycosylation forms of subtype H5 trimeric HA protein (sH5(3)) were produced by expression in insect cells and different mammalian cells in th...

Counterelectrophoresis (CEP) and immunodiffusion (ID) were evaluated prospectively as methods for the early and rapid laboratory diagnosis of California encephalitis (CE). CEP and ID studies were done on paired sera from 127 patients with acute central nervous system infections. After the precipitin tests were completed, conventional hemagglutination-inhibition, neutralizing, and complement fix...

The highly pathogenic avian influenza A (HPAI) viruses of the H5N1 subtype infect poultry and have also been spreading to humans. Although new antiviral drugs and vaccinations can be effective, rapid detection would be more efficient to control the outbreak of infections. In this study, a phage-display library was applied to select antibody fragments for HPAI strain A/Hubei/1/2010. As a result,...
