Abstract:

Background :Gelatin and chitosan are known as biodegradable and biocompatible biopolymers. These biopolymers have recently receivedincreasingly more attention for tissueengineering. The aim of this study was to evaluate the toxicity effects of gelatin-chitosan film on bone marrow stromal cell(BMSCs) culture in rat.                                                  Materials & Methods: First, gelatin- chitosan composites film were prepared by solution mixing, followed by film casting of both biopolymers in glacial acetic acid. After two passage of BMSCs culture, the cells were cultured in four plate groups including: control plates have no any film, gelatinplates, chitosan plates and gelatin- chitosan plates.Theproliferation, differentiation, viability and apoptosis rates of BMSCs were studied duringthesecond, fourthand sixth days. The activity of superoxidedismutase(SOD),catalase(CAT)and  glutathione(GSH) and malondialdehyde(MDA) levels were determined by using biochemical methods. Results:The results showed that the mean BMSCs proliferation significantly reduced in chitosan group compared to control group (p

CFU-GM Like Colonies Derived from Embryonic Stem Cells Cultured on the Bone Marrow Stromal Cells

The aim of this study was to isolate mouse embryonic stem cells from late blastocyst stage embryos and to use them as a model system for the study of hematopoietic induction outside the embryo by coculturing of embryonic stem cells with bone marrow stromal cells. Blastocyst stage embryos from pregnant NMRI mice were obtained and cultured for 1-2 days in DMEM medium. The inner cell masses formed...

Repressing of SOX6 and SOX9 in Situ Chondrogenic Differentiation of Rat Bone Marrow Stromal Cells

Introduction: SOX9 is a transcriptional activator which is necessary for chondrogenesis. SOX6 are closely related to DNA-binding proteins that critically enhance its function. Therefore, to carry out the growth plate chondrocyte differentiation program, SOX9 and SOX6 collaborate genomewide. Chondrocyte differentiation is also known to be promoted by glucocorticoids through unknown molecular mec...

Perspectives of chitosan nanofiber/film scaffolds with bone marrow stromal cells in tissue engineering and wound dressing

Objective (s): Several methods have been proposed for repairing defects and damages, one of which is cell therapy. Bone marrow stromal cells seem to be suitable for this purpose. On the other hand, many biometric materials are used to improve and correct the defects in the body. Nanofibers are widely used in the medical industry, especially in tissue engineering, as scaffolds in wound healing a...
