Abstract:

Background: Due to increased cancer chemotherapy resistance, the use of natural compounds in cancer treatment has been considered in recent years. Inula-Oculus-Christi, is one of the high importance plants in the field of traditional medicine. It has been shown that extract of this plant has cytotoxic effects on breast cancer cells. Objective: Therefore, the aim of this study was to evaluate the cytotoxic effect of Gaillardin and apoptosis induced by this extract on acute lymphoblastic leukemia cells. Methods: Gaillardin from Inula-Oculus-Christi plant was extracted by Traditional Medicine Research Center of Shahid Beheshti University of Medical Sciences. The cytotoxic effect of Gaillardin on the acute lymphoblastic leukemia (Nalm-6) and PBMC (as a normal cell line) was investigated at different doses for 24 and 48 hours. In this study, Trypan blue, MTT assay and DAPI staining were used. In addition, Annexin V/PI staining was utilized for confirming apoptosis. Results: The results of Trypan blue and MTT assay demonstrated the induction of apoptosis and reduction in proliferation of Nalm-6 cells treated by Gaillardin, showing a time and dose-dependent manner (P < 0.001). However, low sensitivity to the different concentrations of Gaillardin was observed in PBMCs. DAPI staining and Flow cytometry analysis confirmed a significantly high percentage of apoptotic cells compared with control groups (P < 0.001). Conclusion: According to the results, Gaillardin demonstrated cytotoxic effects on Nalm-6 cells, but

Investigating the apoptotic and anti-proliferative effects of Juniperus excelsa fractions on Acute Lymphoblastic leukemia cell line (Nalm-6)

Abstract Background and Objectives Acute lymphoblastic leukemia is a hematological malignancy of lymphoid lineage. Side effects of chemotherapy drugs is a major reason for the tendency to use Herbal medicines. Previous studies on Juniperus excelsa (J.excelsa) have shown anti-cancer properties on some cancer cell lines. This study aimed to investigate the effect of three fractions of J.excelsa ...

A Comparison between the Anticancer Activities of Free Paclitaxel and Paclitaxel-Loaded Niosome Nanoparticles on Human Acute Lymphoblastic Leukemia Cell Line Nalm-6

Background: Niosomes or Nonionic surfactant vesicles are nano vehicles utilized in drug delivery systems, especially in cancer therapy. In this study, these vesicles were applied as delivery system for anticancer drug, paclitaxel and then, its anticancer activities was compared with free paclitaxel on Human Acute Lymphoblastic Leukemia (ALL) cell line Nalm-6. Materialas and Methods: In this exp...

Gallic Acid Inhibits Proliferation and Induces Apoptosis in Lymphoblastic Leukemia Cell Line (C121)

AbstractLeukemia is known as the world’s fifth most prevalent cancer. New cytotoxic drugs have created considerable progress in the treatment, but side effects are still the important cause of mortality. Plant derivatives have been recently considered as important sources for the treatment of various diseases, including cancer. Gallic acid (GA) is a polyhydroxyphenolic compound with a wide rang...

Effects of valproic acid and pioglitazone on cell cycle progression and proliferation of T-cell acute lymphoblastic leukemia Jurkat cells

Objective(s): T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematologic malignant tumor. Administration of chemical compounds influencing apoptosis and T cell development has been discussed as promising novel therapeutic strategies. Valproic acid (VPA) as a recently emerged anti-neoplastic histone deacetylase (HDAC) inhibitor and pioglitazone (PGZ) as a high-affinity peroxisome p...

Mir-55 inhibition can reduce cell proliferation and induce apoptosis in Jurkat (Acute T cell Leukemia) cell line

Background MicroRNAs are small and non-coding RNA molecules with approximately 22 nt in length that cause inhibition of translation or degradation of mRNA. MiR-155 is a kind of molecule with different functions, such as its role in proliferation, apoptosis, inflammation, differentiation, and immunity. One of its best known functions is apoptosis that affects on caspase-3 activity. The main aim...
