P-58: Evaluation of Ovarian Toxicity Effects of Chlorpromazine in Female Rats

Abstract:

Background: Elevations in serum prolactin levels (hyperprolactinaemia) are a common side effect of conventional antipsychotic treatments. The main objective of this study was to consider the effects of the antipsychotic drug chlorpromazine (CPZ) in female rats reproductive system and to examine the relationship between the effects on reproductive organs and hormone concentrations. Materials and Methods: In this study fourty adult female wistar rats (160 ± 20g) were randomly divided into five groups (n=8 each) with one group serving as control that received only food and water,and one group serving as control sham((5 mL/kg of 0.5% methylcellulose solution.), in the drug treated groups female rats were orally administered CPZ (3, 10, 30 mg/kg) once daily (7 days/week) for 4 weeks .The test compound was suspended or dissolved in 0.5% methylcellulose solution . All rats were sacrificed by Co2 inhalation one day after the last drog administration .The reproductive organs were removed surgically and prepared for histological study . Results: This study showed that treated by CPZ had significant increase in prolactin level in plasma, because of high activity of prolactin on ovary spesially on luteotropic which cause in the female reproductive organs, enlarged corpora lutea and significantly increased antral and preantral atretic follicles number in the ovary, atrophy in the uterus and epithelial mucification in the vagina were obviuse. Conclusion: These results showed that oral CPZ treatment indused ovarian toxicity including increase in atresia of the large follicles and changed the fertility parameters.
